补脾强力复方对自身免疫性重症肌无力大鼠胸腺组织GR mRNA表达的影响 被引量:9

Effect of Bupi Qiangli Compound on mRNA Expression Level of Glucocorticoid Receptor in Thymus of EAMG Model Rats

摘要 目的:观察补脾强力复方对实验性自身免疫性重症肌无力(EAMG)模型大鼠胸腺组织中糖皮质激素受体(GR)mRNA表达的影响。方法:将Lewis大鼠按体重随机分为6组,即:正常组、模型组、强的松组(5.4 mg/kg)、补脾强力复方不同剂量组(剂量分别为4.75 g/kg,7.12 g/kg,9.49 g/kg),除正常组外,均采用鼠源性AchR-α亚基97-116肽段序列(R97-116)免疫接种复制EAMG模型。药物治疗后,ELISA法检测各组大鼠的AchR-Ab、皮质醇水平,RT-Q-PCR技术检测大鼠胸腺组织GR mRNA表达水平。结果:与正常组比较,模型组GR mRNA相对表达量降低(P<0.01),与模型组比较,西药组、中药大、中、小剂量组大鼠AchR-Ab含量下降(P<0.05),强的松组、中药大剂量组和中剂量组GR mRNA相对表达量增加(P<0.05)。结论:补脾强力复方可提高胸腺组织GR mRNA的表达,这可能是其治疗MG的作用机制之一。 Objective : To observe the effect of Bupi Qiangli Compound on the mRNA expression level of glucocorticoid receptor (GR) in the thymus of experimental autoimmune myasthenia gravis(EAMG) model rats. Methods : Female Lewis rats were random- ly divided into six groups according to the different weight. That is:normal group, model group, Prednisone group (5.4 mg/kg), different dose of Bupi Qiangli Compound groups(4.75 g/kg,7.12 g/kg,9.49 g/kg). All Lewis rats were immunized with 97 - 116 peptide of the rat original AchR α- subunit( R97 -116) to copy EAMG rats model except for normal group. The levels of AchR - Ab and cortisol were determined by ELISA and the mRNA expression levels of GR were determined by RT - Q - PCR af- ter the treatment. Results : Compared to the normal group, the mRNA expression levels of GR decreased in model group ( P 〈 0. 01 ). Compared to the model group, the levels of AchR - Ab decreased in Prednisone group and different dose of Bupi Qiangli compound groups(P 〈 0.05 ),the mRNA expression levels of GR increased in Prednisone group,Bupi Qiangli compound large and middle dose treatment group (P 〈 0.05 ). Conclusion:Bupi Qiangli Compound can increase the mRNA expression level of glu- cocorticoid receptor in thymus, which may be one of the therapeutic mechanism of MG.
