摘要
目的探讨菝葜鞣质抗肺癌活性及其作用机制,为菝葜临床用于治疗肿瘤疾患提供科学实验依据。方法采用细胞计数仪测定给药后肺癌细胞的死亡率、流式细胞仪检测细胞周期分布及细胞凋亡率、Westrn Blot检测Bcl-2、Bax及CyclinD2蛋白的表达水平。结果菝葜鞣质可升高肺癌细胞死亡率、诱导细胞凋亡并使其细胞周期阻滞在G0/G1期;随着给药浓度的增加,Bcl-2、CyclinD2表达水平下降,Bax的表达水平无明显变化。结论菝葜鞣质具有抗肺癌活性,其作用机制可能是通过下调Bcl-2及CyclinD2的表达促进A549细胞凋亡并使其细胞周期阻滞在G0/G1期。
Objective To study the activity and mechanism of tannins from Smilax china L.( SCL) against lung cancer cell. Methods The cell counting instrument was used to detect the death rate of lung cancer cells,flow cytometry was applied to detect the apoptosis and cell cycle of A549 after treatment with tannins of SCL. The expression levels of Bcl-2,Bax and CyclinD2 in A549 cells were determined by Western Blot. Results Tannins from SCL could increase the mortality of lung cancer cell and arrest the cell cycle in G0 / G1 phase. With the increase of drug concentration,the expression level of Bcl-2 and CyclinD2 were down-regulated and there was no obvious change in Bax. Conclusion Tannins from SCL has activity against lung cancer,its mechanism may be related to reducing the Bcl-2 and CyclinD2 expression levels and making the cell cycle arrest in G0 / G1 phase.
