摘要
目的揭示穴贴对幼鼠早期急性牙髓的保护作用,为中医穴贴干预乳牙保健奠定实验基础。方法选取48只1月龄幼鼠,雌雄各半,将幼鼠随机分成空白组、模型组、穴贴颊车组及穴贴阳溪组,每组12只,然后各组再分为穴贴治疗12 h组和穴贴治疗24 h组。采用开髓封内毒素法建立急性牙髓炎模型。各组经穴贴干预后,采用免疫组化法测定幼鼠乳牙Notch1、Notch2蛋白在第12 h、第24 h的表达。结果 12 h各组Notch1蛋白比较,空白组与模型组、穴贴颊车组、穴贴阳溪组比较差异有统计学意义(P<0.05),模型组与穴贴阳溪组比较差异无统计学意义(P>0.05)。24 h各组Notch1表达模型组与空白组、穴贴颊车组、穴贴阳溪组比较差异有统计学意义(P<0.05)。在第12 h各组Notch2蛋白比较,空白组与模型组比较差异无统计学意义(P>0.05),模型组与穴贴各组比较出现有统计学意义(P<0.05);但空白组、穴贴颊连组、穴贴阳溪组24 h Notch2水平与模型组比较差异有统计学意义(P<0.05)。结论 Notch信号蛋白在幼鼠牙髓损伤早期,可通过自身表达增强起到牙髓自我修复作用。Notch1的表达时间早于Notch2,牙穴贴干预后可缩短Notch信号蛋白表达上调时间,提高急性牙髓损伤后的自身修复能力。
Objective To reveal the protective effect of acupoint sticking therapy on immature rats with early acute pulpitis,provide an experimental basis for deciduous teeth health care using acupoint sticking. Methods Fourty-eight immature rats aged one month( sex ratio:1 ∶ 1) were randomly divided into blank group,model group,Jiache acupoint sticking group and Yangxi acupoint sticking group,12 immature rats in each group. The immature rats in the four group were divided into 12-hour subgroup and 24-hour subgroup. Opening pulp chamber and sealing endotoxin( LPS) method was used to establish the model of acute pulpitis. After intervention,immunohistochemical method was used to detect the expression levels of Notch1 protein and Notch2 protein in 12 and 24 hours. Results In 12 hours after intervention,there was statistically significant difference in Notch1 protein expression between blank group and model group,Jiache acupoint sticking group,Yangxi acupoint sticking group( P〈0. 05),but there was no statistically significant difference between model group and Yangxi acupoint sticking group( P〉0. 05). In 24 hours after intervention,there was statistically significant difference in Notch1 protein expression between model group and blank group,Jiache acupoint sticking group,Yangxi acupoint sticking group( P〈0. 05). In 12 hours after intervention,there was no statistically significant difference in Notch2 protein expression between blank group and model group( P〉0. 05),but there was statistically significant difference between model group and Jiache acupoint sticking group,Yangxi acupoint sticking group( P〈0. 05). In24 hours after intervention,there was statistically significant difference in Notch2 protein expression between blank group,Jiache acupoint sticking group,Yangxi acupoint sticking group and model group( P〈0. 05). Conclusion Notch signaling protein can repair dental pulp by enhancing its self expression in early stage. The expression time of Notch1 is earlier than Notch2,the up-regulation time of Notch signal protein expression level is shortened after acupoint sticking therapy,the self-repair ability of dental pulp after damage is improved.
