摘要
目的:观察白鲜皮水提物对小鼠迟发型变态反应的影响。方法取体重为18~22 g的昆明小鼠60只,雌雄各半,按体重随机分6组,即空白对照组、模型对照组、白鲜皮水提物高(11.0 g/kg)、中(55.g/kg)、低(2.8 g/kg)剂量组和阳性对照(地塞米松片2.22 mg/kg)组,每组10只,各给药组分别给予相应浓度的相应药物,空白与模型对照组给予0.015 ml/g 80%吐温20-生理盐水溶液,每日1次,连续给药3 d后,除空白对照外,每只鼠腹部用5%DNCB溶液均匀涂抹进行致敏,第4 d强化一次,于给药后第9 d,将5%DNCB溶液均匀涂抹于小鼠右耳进行攻击,空白对照组同样涂耳但未致敏,攻击后24 h,测定耳肿胀度和小鼠的脾脏指数和胸腺指数。结果以耳肿胀度为指标,与模型组相比较,白鲜皮高剂量组、阳性对照组具有极显著性差异(P<0.01),白鲜皮中剂量组具有显著差异(P<0.05),白鲜皮低剂量组则无显著性差异;以胸腺指数、脾脏指数为指标,与模型组相比较,白鲜皮高剂量组、白鲜皮中剂量组、阳性对照组具有极显著性差异( P<0.01),白鲜皮低剂量组胸腺指数较模型组具有显著性差异( P<0.05)。结论白鲜皮水提物可抑制DNCB诱导的小鼠迟发型变态反应( DTH)及抑制小鼠的特异性细胞免疫功能的作用。
Objective To observe the effect of the cortex dictamni on the delayed type hypersensitivity in mice.Methods 60 Kunming mice weighing 18~22g, half female and half male , were randomly and averagely divided into 6 groups according to weight:blank control group, model control group, the cortex dictamni(11.0g /kg), in (5.5g /kg) and low (2.8g /kg) dose group and positive control group (dexamethasone tablets 2.22mg/kg) , 10 rats in each group , The corresponding concentrations of each drug group were given respectively , the blank and the model control group were given 0.015ml/g 2%Twain 80%saline solution, 1 times a day, after 3 days of continuous administration , in addition to the blank control , each mouse abdomen with 5%DNCB solution was evenly coated for sensitized , fourth days to strengthen once , in ninth days , the 5%DNCB solution evenly attack on mouse ear , the blank control group also painted ear but not sensitized , after the attack of 24h, determination of ear swelling degree and the index thymus and spleen .Results In ear swelling as an indicator , comparing cortex dictamni , high dose group and model group , positive control group with a very significant difference (P〈0.01), cortex dictamni dose group were significantly different (P〈0.05), cortex dictamni leather low-dose group had no significant difference; thymus and spleen indexes as an indicator , as compared with the model group cortex dictamni leather high-dose group , cortex dictamni dose group , positive control group with a very significant difference (P〈0.01), cortex dictamni on the low-dose group compared with the model group , the thymus index had significant difference ( P〈0.05) .Conclusions The cortex dictamni can inhibit DNCB induced murine delayed hypersensitivity ( DTH) and has the effect of inhibiting the specific cellular immune function of mice.
