大鼠肠神经系统神经元的原代培养及鉴定

摘要 目的探讨大鼠ENS神经元原代分离培养及鉴定的方法。方法解剖分离大鼠小肠,显微剥离小肠肌层,消化后,种植于培养板,用无血清培养基换液培养,观察神经元生长情况并进行免疫荧光鉴定。结果成功分离ENS神经元,神经元在无血清培养基中生长良好,呈典型神经元形态,经免疫荧光鉴定证实绝大部分细胞是神经元。结论成功建立了大鼠ENS神经元原代培养和鉴定的方法,为深入研究ENS神经元奠定了基础。 Objective To investigate the primary isolation , culture and identification of rat ENS neurons . Methods The small intestine was dissected from rat and then digested into single cell suspension .The cells were planted onto 24-well plate and cultured with serum-free mediums .The growth of neurons was observed and the neurons were identified with immunocytofluorescence .Results ENS neurons were successfully isolated and survived well in serum-free medium.Immunocytofluorescence showed most of the cultured cells were neurons . Conclusions A method has been established for primary culture and identification of rat ENS neurons .It paves way for further research of ENS neurons .
