摘要
目的:考察七宝美髯口服液对SAMP8小鼠学习记忆功能及海马Neu N、Nampt和Aβ和表达的影响。方法:SAMP8小鼠灌胃给予七宝美髯口服液,SAMR1小鼠平行对照,连续干预60d,采用Morris水迷宫法考察小鼠学习记忆功能,采用免疫组化法考察小鼠海马Neu N、Aβ和Nampt的表达。结果:与模型对照组比较,七宝美髯口服液(1.44g/kg)能显著提高SAMP8小鼠的学习记忆能力,缩短逃避潜伏期,增加小鼠进入平台象限次数,提高进入平台象限百分比;能显著促进海马Neu N和Nampt的表达,提高其平均光密度值;能显著抑制小鼠海马Aβ的表达,降低其平均光密度值。结论:七宝美髯口服液能够改善SAMP8小鼠的学习记忆功能,该作用与其增强SAMP8小鼠海马Neu N和Nampt表达,抑制Aβ的表达有关。
Objective: To investigate the effects of Qibaomeiran oral liquid on learning and memory function and the expression of NeuN, Nampt and Aβ in hippocampus of SAMP8 mice. Methods:Qibaomeiran was administered by oral gavage to SAMP8 for 60 days while SAMPI mice were set as negative control-group. Inspecting learning and memory function by Morris water maze and evaluating the expression of NeuN, Nampt and Aβ in hippocampus by immunohistochemistry. Results: Qibaomeiran oral liquid(1.44g/kg) can not only improve the learning and memory function, shorten escape latency, advance space exploration ability obviously but also increase the times and percentage that SAMP8 mice entered the platform quadrant, additionally,it up-regulated the expression of NeuN and Nampt in hippocampus and increase the average optical density value. Meanwhile, it down-regulated the expression of Aβ, decreased significantly its average optical density value. Conclusion: Qibaomeiran oral liquid can improve the learning and memory function and protect neurons in hippocampus of SAMP8 mice by promoting NeuN, Nampt and weakening Aβ in hippocampus.
