摘要
目的:研究泽泻提取物对非酒精脂肪肝的保护作用及其相关作用机制。方法:将SD大鼠随机分成5组:正常组、模型组及设置400、200、100mg/kg泽泻提取物为高、中、低剂量组,每组10只;正常组以基础饲料喂养及饮水,其余各组给予高脂饲料及饮水;用RT-PCR法检测肝组织内CYP2E1、CYP2A5基因的表达情况,用ELISA法检测血清中SOB、AST、ALT、TG、HDL、LDL的表达情况,称量各组老鼠前后体重。结果:与模型组相比,泽泻提取物剂量组肝组织AST、ALT、TG、LDL、CYP2E1、GYP2A5及体重均有所降低,SOD、HDL升高。结论:泽泻提取物可能通过降低肝组织中AST、ALT、TG、LDL、CYP2E1、CYP2A5的表达,升高SOC、HDL而保护非酒精脂肪肝。
Objective: To study the protective effect ofAlisma orientalis extract on nonalcoholic fatty liver and its related mechanism. Methods: SD rats were randomly divided into 5 groups, normal group and model group. 400, 200, 100 mg / kg Alisma orientalis extracts were divided into high, middle and low dose group. The normal group was fed with basic feed and drinking water, and the other groups were given high fat diet and drinking water. The expression of CYP2E 1 and CYP2A5 in liver tissue were detected by RT-PCR. The levels of SOD, AST, ALT, TG, HDL and LDL in serum were measured by ELISA. The body weight. Results: Compared with the model group, the AST, ALT, TG, LDL, CYP2E1, CYP2A5 and body weight of the alisma extract decreased, SOD and HDL increased. Conclusions : Alisma orientalis extract could protect the nonalcoholic fatty liver by elevating the levels ofAST, ALT, TG, LDL, CYP2E 1, CYP2A5, SOD and HDL.
