摘要
目的:探讨四神丸及其拆方(去补骨脂)对脾肾阳虚泄泻大鼠瘦素(Leptin)、胆囊收缩素(CCK)和三磷酸肌醇(IP3)的影响。方法:将50只SD大鼠随机分为正常组、模型组、得舒特组、四神丸组及四神丸拆方组各10只;采用灌服大黄水煎液和肌内注射氢化可的松的方法复制脾肾阳虚泄泻大鼠模型,灌胃14 d后观察各组大鼠的一般状态,检测血清中Leptin、CCK和胃窦、十二指肠组织中IP3含量变化。结果:模型组大鼠的体质量、血清Leptin、CCK含量均低于正常组,胃肠组织IP3含量高于正常组,差异均有统计学意义(P<0.01)。给药后四神丸组、四神丸拆方组和得舒特组体质量、血清Leptin、CCK含量均高于模型组,IP3均低于模型组,差异均有统计学意义(P<0.05或P<0.01)。四神丸组的体质量高于得舒特组和四神丸拆方组,血清CCK含量高于得舒特组,十二指肠IP3含量低于四神丸拆方组,差异均有统计学意义(P<0.05);四神丸拆方组胃肠组织中IP3含量高于得舒特组(P<0.05)。结论:四神丸和四神丸拆方可增加脾肾阳虚泄泻大鼠血清Leptin、CCK含量,降低胃肠中IP3含量;四神丸及其拆方对脾肾阳虚泄泻大鼠的作用可能与调节Leptin、CCK、IP3水平有关。
Objective: To explore the effect of Sishenwan and its disassembled prescriptions (get rid of Fructus Psoraleae) on Leptin, CCK and IP3 of rats with diarrhea of Asdthenic Splenonephro-yang (AS). Methods: The 50 SD rats were randomly divided into normal group, model group, Dicetel group, Sishenwan group and its disassembled prescriptions group, 10 rats iu each. The model was copied by gavage rhubarb decoction and intra- muscular injection of hydrocortisone. The general state, Leptin and CCK content in serum, IP3 in the gastric antrum and duodenum were observed after 14d. Results: The model group showed lower body weight loss, serum Leptin and CCK than the normal group, while higher IP3 in gastrointestinal tissue than the normal group (P〈0.O1). The Sishenwan group and its disassembled prescriptions group and Dieetel group showed higher body weight loss, serum Leptin and CCK than the normal group, while lower IP3 in gastrointestinal tissue than the normal group, with statistically significant difference (P〈0.05 or P〈0.01). The Sishenwan group showed higher body weight than Dicetel group and disassembled prescriptions group, higher serum CCK than Dicetel group, and low- er IP3 content in the duodenum than disassembled prescriptions group. There were statistically significant differ- ences (P〈0.05). The disassembled prescriptions group showed higher IP3 content in gastrointestinal tissue than Dicetel group (P〈0.05). Conclusion: The effect of Sishenwan and its disassembled prescriptions to rats with diar- rhea of AS may be related to the regulation of Leptin, CCK, and IP3 levels.
