摘要
目的探讨轮状病毒对乳鼠肠道外损害及Cp G ODN的保护作用。方法 24只肠道外轮状病毒感染乳鼠随机平均分为3组:对照组(生理盐水处理)、感染组(腹腔注射轮状病毒)、Cp G ODN组(连续腹腔注射50μg Cp G ODN后腹腔注射轮状病毒悬液);处理前后检查各组心、肝、肺组织改变,HRV VP7 mRNA水平以及心肝功能。结果感染组乳鼠心、肝、肺组织出现严重病理学改变,且心、肝组织HRV VP7 mRNA呈阳性,肺组织呈阴性。Cp G ODN组乳鼠心、肝、肺组织病理学改变较轻,心、肝组织HRV VP7 mRNA为弱阳性,肺组织为阴性。Cp G ODN组乳鼠FS、EF显著高于感染组但显著低于对照组,ALT、AST和LVESD显著低于感染组但显著高于对照组差异均有统计学意义(P<0.05)。结论腹腔注射轮状病毒损伤了乳鼠心、肝、肺组织及心肝功能;Cp G ODN预处理减轻了轮状病毒的损伤,对乳鼠具有保护作用。
Objective To observe the extraintestinal injury of neonatal mice infected by rotavirus and the influence of Cp G ODN intervention.Methods 24 extraintestinal rotavirus infected neonatal mice were selected and randomly divided into the control group(physiological saline treatment),the infection group(intraperitoneal injection of HRV suspension) and Cp G ODN group(intraperitoneal injection of HRV suspension after Cp G ODN intervention).Before and after treatment,organizational changes,HRV VP7 mRNA levels,liver function and heart function of the 3 groups were tested.Results Heart,liver,and lung tissue of infection group had severe changes,and HRV VP7 mRNA in heart and liver tissues were positive,while lung tissues were negative.After Cp G ODN intervention,heart,liver,and lung tissue of neonatal mice infected rotavirus had minor changes.HRV VP7 mRNA of heart and liver tissue was weak positive,but that was week negative in lung tissue.After Cp G ODN intervention,FS and EF of Cp G ODN group were significantly lower than control group,but ALT,AST and LVESD of Cp G ODN group were significantly lower than infection group,but significantly higher than control group(P〈0.05).Conclusion Intraperitoneal injection of rotavirus can injure lung,heart,and liver tissue,heart function,and liver function; Cp G ODN intervention can lighten the rotavirus injury and protect neonatal mice.
