摘要
为了研究常用抗生素对临床分离到的鸭疫里默氏杆菌(riemerella anatipestifer,RA)的抑菌效果,试验首先绘制RA的生长曲线,确定其对数生长期,以对数生长期的细菌作为受试菌,测定6种常用抗生素对4株RA菌株在不同稀释度时的最小抑菌浓度(MIC),确定最佳接菌量,进而确定测定MIC的最佳时间。结果表明:新鲜培养的RA菌液的OD600值为0.8~1.2时,细菌达到对数生长期,菌落形成单位达到1×10^(11)~1×10^(12)cfu/m L。对数生长期细菌稀释至1×10^(-5)为最佳接菌量,将上述菌液接种到96孔板后,在37℃培养22~24 h为检测MIC的最佳时间。
To study the antimicrobial effects of commonly used antibiotics on Riemerella anatipestifer (RA) isolated from the clinic, the growth curve of RA was plotted to determine the bacterial logarithmic growth phase at first. The minimal inhibitory concentrations (MICs) of six com- monly used antibiotics against four RA strains at different dilutions were determined using the bacteria in logarithmic growth phase as test bacteria, and the optimal inoculation amount of bacteria and the optimal time for determining MIC was ascertained, respectively. The results showed that the bacteria reached the logarithmic phase when the ODe00 values of bacterial liquid of freshly cultured RA strains were 0.8 to 1.2, and the colony - forming unit (cfu) reached from 1 ×10^11 cfu/mL to 1 ×10^12 cfu/mL. The results indicate that the optimal inoculation amount of bacteria is obtained when the bacteria in logarithmic growth phase are diluted to 1 ×10^ -5, and the optimal time for detecting MIC is obtained when the bacteria cultured at 37℃ for 22 h to 24 h after the bacterial liquid are inoculated into 96 - well plate.
