摘要
目的探讨两色金鸡菊提取物对糖尿病大鼠血管内皮生长因子(VEGF)和细胞间黏附分子1(ICAM1)表达的影响。方法采用高糖、高脂喂养联合腹腔注射链脲佐菌素(streptozotocin,STZ)建立2型糖尿病SD大鼠模型,成模大鼠随机分为正常组、模型组、阳性药二甲双胍组及两色金鸡菊乙酸乙酯提取物高、中、低剂量组共6组,每组10只。给药4w后,采用免疫组织化学染色法检测各组大鼠视网膜中VEGF和ICAM1蛋白的表达。结果与正常组比较,模型组大鼠视网膜VEGF和ICAM表达水平明显提高(P〈0.05);与糖尿病模型组比较,两色金鸡菊乙酸乙酯提取物中剂量组糖尿病大鼠视网膜VEGF和ICAM1的表达均明显降低(P〈0.05)。结论两色金鸡菊乙酸乙酯提取物可通过下调VEGF和ICAM1的表达,抑制糖尿病大鼠视网膜血管新生和炎症反应,发挥对糖尿病视网膜损伤的保护作用。
Objective To investigate the effect of Ethyl acetate extract of Coreopsis tinctoria Nutt on vascular endothelial growth factor (VEGF)and intercellular adhesion molecule 1 (ICAM1 )in diabetic rats. Methods Type 2 diabetic SD rat model was set by high-sucrose and high-fat diet combined with intraperitoneal injection of streptozotocin (streptozotocin,STZ),the rats were randomly divided into normal group,model group,Coreopsis extract high,middle and low dose group,positive drug metformin group, ten rats for each group.After treatment for 4 weeks,retinal VEGF and ICAM1 protein expression was tested by immunohistochemical staining.Results Compared with the normal group,the expression level of VEGF and ICAM1 in retina of rats of model group was increased significantly (P 〈0.05);Compared with diabetic model group,the expression of VEGF and ICAM1 in diabetic rat retina of Coreopsis extract middle dose group were significantly decreased (P〈0.05).Conclusion Ethyl acetate extract of Coreopsis tinctoria Nutt can inhibit the retinal neovascularization and inflammatory reaction in diabetic rats by decreasing the expression of VEGF and ICAM1,and play a protective role in diabetic retinopathy.
