静脉注射全氟化碳预处理与后处理对急性肺损伤大鼠的影响 被引量:3

摘要 目的探讨静脉注射全氟化碳(PFC)预处理与后处理对脂多糖(LPS)致急性肺损伤大鼠的保护作用及相关作用机制。方法选取24只雄性Wistar大鼠分为对照组(NS组)、LPS组、PFC预处理组(Pre组)和PFC后处理组(Post组)。NS组均以等量生理盐水作对照,LPS组经气管内滴注LPS;Pre组于气管内滴注LPS前经股静脉注射PFC;Post组于气管内滴注LPS后经股静脉注射PFC。NS组于气管内滴注生理盐水后6h,LPS组、Pre组与Post组分别于滴注LPS后6h处死动物。比较各组大鼠肺病理学变化,PaO_2,肺湿干比(W/D),肺髓过氧化物酶(MPO)及细胞黏附分子-1(ICAM-1)表达情况。结果应用PFC明显升高PaO_2,降低肺W/D比值、MPO活性及ICAM-1表达水平(P<0.05),而且Pre组较Post组作用更显著。结论静脉注射PFC对ALI具有保护作用,以PFC预处理效果更为显著,其机制可能与下调ICAM-1表达有关。 Objective To investigate the protective effect and the possible mechanism of intravenous infusion with perfluorocarbon pretreatment and posttreatment on acute lung injury in rats on LPS-induced acute lung injury in rats.Methods A total of 24 Wistar rats were randomly divided into 4groups:control group(NS group),LPS group,Pre group and Post group.Normal saline was given to NS group as a control.Rats were treated with LPS by intratracheal instillation in LPS group,rats received PFC through femoral vein prior to LPS instillation in Pre group and rats received PFC through femoral vein after LPS instillation in Post group.NS group were sacrificed at 6hafter being injected with NS,LPS group,Pre group and Post group were sacrificed at 6hafter being given LPS.Pathological changes of lung,PaO2,lung wet to dry weight ratio(W/D),expression of myeloperoxidase(MPO)and intercellular adhesion molecule-1(ICAM-1)of lung were assessed.Results Intravenous infusion with perfluorocarbons increased PaO2,decreased W/D,MPO and ICAM-1significantly(P〈0.05),and this effect is more remarkable in Pre group.Conclusion Intravenous infusion of PFC significantly protects lung from acute lung injury,especially by pretreatment forms,probably by down-regulate the expression of ICAM-1.
