摘要
以云南永仁野生黄牛肝菌为原料,采用响应面法对微波辅助提取野生菌多糖的工艺进行研究。在单因素实验基础上,选择料液比、水浴温度、水浴时间进行3因素3水平中心组合设计实验,再利用响应面设计法优化黄牛肝菌多糖的提取工艺,并测定了黄牛肝菌多糖对1,1-二苯基-2-三硝基苯肼(DPPH)自由基和羟自由基(OH·)的清除作用。结果表明:黄牛肝菌多糖的最佳提取工艺:水浴温度78℃,液料比52∶1(m L/g),水浴时间2.60 h,微波时间70 s,微波功率350 W,在此条件下多糖平均得率为14.74%,与预测值(14.77%)相比,相对误差较小,为0.35%。黄牛肝菌多糖对DPPH自由基及OH·均有一定的清除作用,且随其随浓度的增大而增大。
In this research ,microwave-assisted method was used to extract polysaccharides from Boletus auripes Pk.of Yongren area in Yunnan and this technology was optimized by response surface method.Based on single-factor experiment,three-factor and three- level central composite design was conducted with certain liquid - solid ratio, water bath temperature and time. Meanwhile,its scavenging effect on DPPH free radical and OH. was investigated.The optimal extraction processes as follows: liquid-solid ratio at 52:1 (mL/g) , water: bath temperature of 78℃ , water bath time for 2.60 h, microwave time in 70 s and microwave power at 350 W. It was found that polysaecharide yielded was up to 14.74% compared with theoretical value ( 14.77 % ) , the relative error was at 0.35 %. Furthermore, it had certain scavenging effect on DPPH free radical and OH·, which became larger with the increase of its concentration.
