COM.TEC型血细胞分离机采集外周血单个核细胞效果分析 被引量:1

摘要 目的观察费森尤斯COM.TEC型血细胞分离机autoMNC程序采集患者外周血单个核细胞的效果。方法粒细胞集落刺激因子动员后,采用COM.TEC型血细胞分离机autoMNC程序采集患者外周血单个核细胞,流式细胞仪检测CD34^+细胞和单个核细胞百分比,血常规检测白细胞计数。对外周血单个核细胞采集效率与单个核细胞采集参数及采集物质量指标进行相关性分析。结果 28例患者平均处理血量2.34±1.75L,采集物体积46.63±28.17mL,白细胞计数(11.40±10.40)×10^(10)个细胞/L,单个核细胞百分比(83.79%±21.77)%,CD34^+细胞百分比(0.81±1.54)%。MNCs采集效率与外周血处理血量呈正相关(r_s=0.538,P<0.05),和采集前外周血MNCs计数呈负相关(r_s=-0.379.P<0.05);MNCs采集效率与采集物白细胞和单个核细胞总数呈正相关(r_s=0.866,r_s=0.924,P<0.05),与采集前外周血白细胞计数、采集物CD34^+细胞和单个核细胞百分比之间无相关性(P>0.05)。结论外周血单个核细胞采集效率作为评价COM.TEC型血细胞分离机autoMNC程序采集效果的指标,可综合反应重要的采集参数与采集物质量。 Objective To observe the efficiency of Fresenius COM.TEC blood cell separator autoMNC program on collection of peripheral blood mononuclear cells(MNCs).Methods After mobilization with granulocyte colony- stimulating factor,the peripheral blood MNCs were collected by autoMNC program of Fresenius COM.TEC blood cell separator.The percentage of MNCs and CD34+ cells were detected by flow cytometry.The white blood cells(WBC) were counted by blood routine.The correlation analysis were performed between collection efficiency(CE) of MNCs and the count of MNCs and WBC in peripheral blood,total WBC,the percentage of MNCs and CD34+ cells in collection.Results The processed mean blood volumes were 2.34±1.75 L in 28 patients.Collection volume was 46.63±28.17 ml.The count of WBC was(11.40±10.40) ×10^10cells/L in collection.The percentage of MNCs and CD34+ cell were(83.79±21.77)%and(0.81±1.54)%in collection,respectively.There was a positive correlation between the CE of MNCs and prcessed blood volume(rs=0.538,P〈0.05).But a negative correlation between the CE of MNCs and the count of MNCs in peripheral blood before collection(rs=-0.379,P〈0.05).A positive correlation between the CE of MNCs and the count of WBC and MNCs in collection(rs was 0.866 and 0.924,P〈0.05).There were no correlations between the CE of MNCs and count of WBC in peripheral blood before collection percentage of CD34+ cells and MNCs in collection(P〉0.05).Conclusion Collection efficacy of peripheral blood MNCs using COM.TE blood cell separator autoMNC program could reflect the important collection parameters and the quality of the collection.
