摘要
为了快速检测战场水环境中TNT含量以判断水源可用性,提出了建立关于TNT的酶联免疫吸附分析法(ELISA)。分析ELISA的建立要点:根据TNT分子结构官能团的可改造性提出了合成半抗原的3种方法;对应3类半抗原采用不同蛋白偶联方法合成3种人工抗原;分析动物体对TNT人工抗原可产生有效免疫应答并可获得满足ELISA要求的抗血清;样品前处理方法可满足快速检测要求。从理论证明建立TNT的ELISA并满足检测要求的可行性;同时,提出了利用此法检测待解决和优化的问题。
To determine the availability of water sources by rapidly detecting the TNT concentration in battlefield water environment,the enzyme-linked immunosorbent assay of TNT was put forward. The key points of ELISA method were analyzed as below:three kinds of semi-antigen methods were proposed based on modification of TNT functional groups. Three artificial antigens were synthesized by employing different proteins coupling reactions corresponding to three kinds of semi-antigens. Artificial antigens of TNT stimulated animals to produce effective immune responses and produced antiserum to comply with requirements of ELISA. Sample pretreatment method was suitable for the rapid detection. Above research theoretically proved that the ELISA method was feasible and satisfactory for detecting TNT. Meanwhile,some problems urgently needed to be solved and optimized by the method were put forward.
