摘要
目的论关于眶额叶区5-HT以及Glu表达水平与脑梗死后抑郁症关联。方法健康成年雄性大鼠进行实验,饲养1周后造模,造模组动物6只;同时选取对照6只,进行实验研究。借助高效液相色谱实验进行5-HT和多巴胺的含量测定;Glu含量检测首先制备标准品后制备样品,进样条件:流速:1.0 m L/min,检测波长为360 nm。实验结束后,麻醉乌拉坦麻醉实验动物后微量注射澳酚兰溶液后处死取脑,多聚甲醛固定,冰冻切片机作连续的冠状切片,镜检染液的组织学部位。统计分析采用SPSS19.0统计软件包完成,结果效液相色谱分析,测定5-HT标准品溶液峰面积,出峰时间为7.367 min,绘制标准曲线,其线性范围为2~300 ng/m L;建模组与对照组5-HT含量测定结果提示,造模组5-HT表达水平为(341.89±36.98)ng/g,对照组为(335.77±50.89)ng/g,两组水平未见显著差别(P>0.05)。进一步测定不同浓度下的Glu标准品溶液峰面积,出峰时间为11.231 min,线性范围14.1~142μg/m L;各组Glu含量存在差别,造模组高于对照组水平,两组Glu水平分别为(1352.34±431.22)μg/g和(976.25±85.66)μg/g,(P<0.01)。最后,造模组与对照组5-HT表达未见明显差别,继续通过给与外源性注射5-HT 1A受体激动剂8-OH-DPAT后观察,发现大鼠的抑郁样行为得到显著改善,主要表现为糖水偏爱率的提高等方面。结论脑卒总后抑郁样行为的发生可能与眶额叶5-HT水平存在关联,但并不因其直接引发,而可能是通过Glu能系统的介导后产生的,该调节作用由1A受体介导,Glu能系统异常会导致5-HT抗抑郁的效果受影响或无法实现。
Objective To discuss the association of 5-HT and Glu expression levels with post-infarction depression in the orbital frontal lobe. Methods 12 healthy male rats were randomly divided into two groups. One animal model was established. The contents of 5-HT and dopamine were determined by high performance liquid chromatography (HPLC). Glu content was measured. The standard sample was prepared after the sample preparation. The flow rate was 1.0 mL/min and the detection wavelength was 360nm. At the end of the experiment,anesthetized rats were anesthetized by microanalysis after injection of urethane solution. The brain sections were fixed with Para formaldehyde and frozen sections were taken for continuous coronal sections. Statistical analysis using SPSS19.0 statistical software package to complete. Results The standard curve was obtained by HPLC and the peak area of 5- HT standard solution was 7.367 rain. The calibration curve was linear in the range of 2-300 ng/mL. The content of 5-HT in the mod- el group and the control group The results showed that the expression level of 5-HT was 341.89±36.98 ng/g in the model group and 335.77±50.89 ng/g in the control group. There was no significant difference between the two groups (P〉0.05). The peak area of Glu standard solution at different concentrations was 11.231 min,and the linear range was 14.1-142 μg/mL. The Glu content in each group was higher than that in the control group. The Glu levels in the two groups were 1352.34±431.22μg/g and 976.25±85.66μg/ g. Finally, there was no significant difference in the 5-HT expression between the model group and the control group. After observing 8-OH-DPAT, the depressive-like behavior of rats was found to be markedly improved by administration of exogenous 5-HT 1A recep- tor agonist hnprovement,mainly for sugar water preference rate and so on. Conclusion Depression-like behavior may be related to the level of 5-HT in the orbital frontal lobe,but not by direct priming,but may be mediated by the Glu energy system. This regulation is mediated by 1A Receptor-mediated,Glu energy system abnormalities can lead to 5-HT antidepressant effects affected or can not be achieved.
