板蓝根组酸的分离与GC-MS分析和体外抗肿瘤活性研究 被引量:2

摘要 目的:从板蓝根中分离板蓝根组酸,用GC-MS进行分析其结构,研究板蓝根组酸对不同肿瘤细胞的体外抑制作用。方法:采用石油醚提取,柱层析分离方法纯化;板蓝根组酸经过甲酯化,运用GC-MS联用技术进行结构研究;体外培养人肝癌细胞BEL-7404、人鼻咽癌细胞CNE-2和人肺癌细胞H-460,用不同浓度的板蓝根组酸分别对细胞作用后,采用MTT法测定药物对细胞的抑制作用。结果:成功提取和分离板蓝根组酸;板蓝根组酸主要由碳原子数为16~18的一个高级饱和脂肪酸和四个高级不饱和脂肪酸的化合物组成;板蓝根组酸能抑制BEL-7404细胞的增殖,其IC50为93.79μg·mL^(-1)。对人鼻咽癌细胞CNE-2和人肺癌细胞H-460有一定的抑制作用,但是不与药物剂量呈正相关性。结论:GC-MS法能对板蓝根组酸进行结构鉴定;板蓝根组酸具有体外抗肿瘤活性。 Objective To isolate the Banlangen-acids from Banlangen and study its structures with GC-MS; To investigate the anti-proliferation effect of Banlangen-acids on the cancer cells. Methods Banlangen-acids were extracted by petroleum ether. The isolation and purification were subjected to silica gel column chromatography. And the structures were study by GC-MS technology after it were esterified with Methanol; The liver cancer BEL-7404 cells and nasopharyngeal carcinoma CNE-2 cells and hmg cancer H-460 cells were cultured in vitro, The cancer cells was cultured in media with Banlangen-acids, The viability of cells was measured by MTT. Results Banlangen-acids were extracted successfully. It contained one higher saturated fatty acid and four higher unsaturated fatty acids and there were 16 to 18 carbon atoms for each compound. Banlangen-acids could exhibited the strong effect against the growth of BEL-7404 tumor cells and showed a dose-dependent manner, the IC50 was 93.79 μg·mL^-1; Banlangen-acids also has inhibitor:,, the growth of CNE-2 cells and H-460 cells, but the effect had not positive correlation with concentration. Conclusions The chemical structures of Banlangen-acids can be elucidated by GC-MS; Banlangen-acids has anti-tumor activity in vitro.
