摘要
目的:研究莪术乙酸乙酯提取物中的化学成分,探讨莪术活性成分对肺癌细胞增殖和迁移能力的影响。方法:乙酸乙酯试剂提取莪术、减压浓缩得到莪术提取物,采用硅胶柱色谱分离方法对提取物进行分离,所得单体成分结合MS、^(13)C-NMR、~1H-NMR,并查阅相关文献进行结构鉴定。MTT法和划痕实验检测莪术活性成分对肺癌细胞的增殖和迁移能力的影响。结果:从莪术乙酸乙酯提取物中分离得到1个倍半萜类单体化合物,鉴定为(5R,6R,7a S)-7a-羟基-5-异丙烯基-3,6-二甲基-6-乙烯基-5,6,7,7a-四氢-4H-苯并呋喃-2-酮(化合物1)。MTT法提示化合物1可抑制PC-9细胞的生长,细胞划痕实验发现化合物1作用的PC-9细胞的迁移能力相比于对照组降低。结论:从莪术乙酸乙酯提取物中分离鉴定得到单体化合物(5R,6R,7aS)-7a-羟基-5-异丙烯基-3,6-二甲基-6-乙烯基-5,6,7,7a-四氢-4H-苯并呋喃-2-酮,且其对PC-9细胞增殖和迁移能力有抑制作用。
Objective: To investigate constituents from Rhizoma Curcumae extracts, and study their effects on proliferation, migration of PC-9 cells. Methods: The Rhizoma Curcumae extracts were obtained through ethyl acetate extraction and vacuum concentration. They were then isolated by silica gel column chromatography method. The structure of composition was confirmed by various spectral methods, including MS, ^13C-NMR,^1H-NMR data. MTr assay and scratch assay were used to evaluate the effect of constituents on cell proliferation and cell migration. Results: One monomer component belonging to sesquiterpenes was isolated from Rhizoma Curcumae and identified as (5R, 6R, 7aS)-Ta-hydroxy-5-isopropenyl-3, 6- dimethyl-6-vinyl-5, 6, 7, 7a-tetrahydro-4H-benzofuran-2-one( compound 1 ). MTT assay showed that the growth of PC-9 cells which were treated with compound 1 was inhibited. Scratch assay demonstrated that cell migration was reduced when the PC-9 cells were treated with compound 1. Conclusion: Compound 1 isolated from ethyl acetate portion was identified as (5R, 6R, 7aS)-Ta-hydroxy-5-isopropenyl-3, 6-dimcthyl-6-vinyl-5, 6, 7, 7a-tetrahydro-4H-benzofuran-2-one. Compound 1 has an effect on PC-9 cells and inhibits the cell proliferation and cell migration.
