摘要
目的探讨丙烯腈(ACN)对大鼠血清中性激素水平影响的动态变化。方法将40只成年雄性健康的SPF级SD大鼠,按体质量随机分为对照组(玉米油)、低、中、高剂量组(ACN染毒剂量分别为12.5、25.0、50.0mg/kg),对各组大鼠分别给予玉米油和不同剂量的ACN灌胃,1次/d,每次5ml/kg,连续12周,每两周心脏采血1次,共7次。将血样离心后取血清,用放射免疫分析法(RIA)测定睾丸(T)、间质细胞刺激素(ICSH)、促卵泡激素(FSH)水平。结果染毒后第4、6、10周,低、中、高剂量组T水平均低于对照组,差异均有统计学意义(P<0.05);染毒后第2和第12周,中、高剂量组的ICSH水平均明显低于对照组和低剂量组,差异有统计学意义(P<0.05);染毒后第2周,高剂量组FSH水平明显高于低、中剂量组;第6周中、高剂量组的FSH水平均低于对照组;第8周高剂量组的FSH水平低于对照组。ICSH时间与染毒组间有交互作用。结论在该实验条件下,随着ACN染毒剂量的增加,血清中T、ICSH和FSH水平在一定时间内有下降,但非某一时间点的直线下降,呈一定的曲线波动。
Objective To explore dynamic effects of acrylonitrile(ACN)on sex hormone in serum of rats.Methods According to the body weight,40 adult specific pathogen free Sprague-Dawley male rats were randomly divided into one control group and three exposed groups(each group of 10).Rats in the exposed group were treated with ACN at dose of 12.5mg/kg,25mg/kg and 50mg/kg respectively once a day for twelve weeks via gavage,the control group were treated with corn oil in the same condition.Heart blood was sampled once two weeks.The levels of testosterone(T),interstitial cell stimulating hormone(ICSH)and follicle-stimulating hormone(FSH)in rat's serum were measured with radioimmunoassay(RIA).Results T in the all exposed groups were significantly lower than the control group at 4,6,10 weeks after exposure(P〈0.05).ICSH level in the high-dose group was significantly lower than the control group and the low dose group at 2and 12 weeks after exposure(P〈0.05).FSH level in the high-dose group was significantly higher than the low-dose group and the middle-dose groups at second week(P〈0.05).FSH levels in the middle-dose group and in the high-dose group were significantly lower than those in the control group at sixth week(P〈0.05).FSH level in the high-dose group was significantly lower than the low-dose group at eighth week(P〈0.05).There was interaction between the exposure dose and the exposure time only in ICSH.Conclusions With the increasing of ACN dose,the level of T,ICSH and FSH have a downward trend among certain times.The downward trend fluctuated to drop like a curve but not a direct line.
