摘要
目的研究烟根中活性化合物芦丁和绿原酸的分离方法及含量。方法烟根乙醇-水提取物浓缩后经乙酯萃取得乙酯组分和水层浓缩液,水层浓缩液经大孔吸附树脂柱层析得90%乙醇组分和50%乙醇组分。使用液质联用(LCMS)技术分析以上三个组分化学成分,利用硅胶、MCI GEL CHP 20P等柱色谱对有效成分进行分离,利用HPLC测定有效成分的含量。结果烟根50%乙醇组分含有芦丁和绿原酸,90%乙醇组分仅含有芦丁,乙酯组分两者皆无;芦丁和绿原酸在烟根中总含量分别为151mg/kg和2.67mg/kg。结论芦丁和绿原酸的极性较大,存在于大极性的组分中。
Objective To investigate the isolation method and the content of active compounds of rutin and chlorogenic acid in Nicotiana tabacum roots. Methods The concentrated solution of the roots of Nicotiana tabacum extracted with EtOH-H2 O were partitioned between EtOAc and H2 O.The extract liquid of EtOAc was evaporated to dryness under low pressure to afford the EtOAc fraction.The extract liquid of H2 O was subjected to D101 macroporous adsorption resin column chromatography(CC)to produce the EtOH-H2O(90∶10)fraction and the EtOH-H2O(50∶50)fraction.The chemical constituents of the above three fractions were analyzed by LC-MS,the active compounds were isolated by silica gel and MCI GEL CHP 20 Pcolumn chromatography methods and the contents of active compounds were determined by HPLC method. Results In the roots of Nicotiana tabacum,the EtOH-H2O(50∶50)fraction contained rutin and chlorogenic acid,the EtOH-H2O(90∶10)fraction only contained rutin,and the EtOAc fraction did not contain these two compounds.The total content of rutin and chlorogenic acid were 151mg/kg and 2.67mg/kg in the roots of Nicotiana tabacum,respectively.Conclusion The polarities of rutin and chlorogenic acid are fairly strong,only exist in high polar fractions.
