摘要
目的:研究生姜醇提取物对肝脏缺血再灌注大鼠氧化应激损伤的保护作用。方法:将120只SD大鼠随机分为6组:假手术组、模型组、生姜醇提取物(100、200、400 mg/kg)组和金纳多;术前2周开始灌胃给药,每天1次;采用夹闭肝门静脉和肝动脉1 h后松夹的方法制备肝脏缺血再灌注大鼠模型。再灌注2 h后,测定肝脏组织中抗氧化酶(SOD、GSH-Px、CAT)活性和丙二醛(MDA)含量;测定血清中总抗氧化能力(T-AOC)水平和转氨酶(ALT、AST)、乳酸脱氢酶(LDH)含量;HE染色法观察肝脏组织形态结构改变;TUNEL法观察肝细胞凋亡状况并计算凋亡指数(Apoptosis Index,AI);Western blotting法检测肝组织中NF-κB表达并进行半定量分析。结果:生姜醇提取物(200、400 mg/kg)组大鼠肝脏组织中SOD和CAT活性较模型组显著升高,而MDA含量和血清中ALT、AST、LDH水平显著降低,其中400mg/kg组GSH-Px活性和T-AOC水平均显著升高;生姜醇提取物各组大鼠肝脏组织形态结构改变和肝细胞凋亡状况较模型组均明显改善,均以400 mg/kg组效果最为显著;生姜醇提取物(200、400mg/kg)组肝细胞AI较模型组显著降低、NF-κB蛋白表达显著下调;上述差异均具有统计学意义(P<0.05,P<0.01)。结论:生姜醇提取物能够有效改善肝脏组织中抗氧化酶活性、抑制肝脏组织病变、改善肝功能、抑制肝细胞凋亡,提示生姜醇提取物对肝脏缺血再灌注大鼠氧化应激损伤具有保护作用。
Objective: To investigate the protective effect of ethanol extract of zingiber officinale on oxidative stress after hepatic ischemia- reperfusion in rats. Methods: 120 rats were randomly devided into six groups:sham operation group,model group,ethanol extract of zingiber officinale 100,200,400 mg / kg groups and Ginaton 4mg / kg group. Two weeks before operation,the drugs were given by intragastric administration,once a day. Two hours after reperfusion,the activity of SOD,GSH- Px,CAT and the content of MDA in hepar were determined; the level of T- AOC and the content of ALT,AST,LDH in serum were determined; the histopathological changes and hepatocyte apoptosis were observed,and the apoptosis index( AI) was calculated; the expression of NF- κB protein was detected by Western blotting and semi- quantitative analysis. Results: Compared with the model group,the activities of SOD,CAT in hepar of ethanol extract of zingiber officinale 200,400 mg / kg groups were significantly increased; the contents of MDA in hepar and the content of ALT,AST,LDH in serum were significantly decreased; the activity of GSH- Px and the level of T- AOC in400 mg / kg group were significantly increased; the histopathological changes and the hepatocyte apoptosis were significantly improved compared with those of model group; the AI of 200,400 mg / kg groups was significantly decreased( P〈0. 01); the expression of NF- κB proten was significantly down-regulated. All of the above differences were significant( P〈0. 05,P〈0. 01). Conclusion: Ethanol extract of zingiber officinale can effectively improve the activity of antioxidase,inhibit the histopathological changes,improve hepar function,depress hepatocyte apoptosis,suggesting that ethanol extract of zingiber officinale has a protective effect on oxidative stress after focal hepatic ischemia-reperfusion in rats.
