SRAP和EST-SSR标记对菊苣遗传多样性的比较分析 被引量:1

Comparative analysis of genetic diversity of Cichorium intybus L. with SRAP and EST-SSR markers

摘要 为了研究中国、意大利、法国、美国等15个国家80份菊苣属种质的遗传多样性及亲缘关系,试验采用SRAP和EST-SSR两种分子标记进行检测。结果表明:SRAP和EST-SSR分别扩增出257条和218条条带,平均多态位点百分率分别为96.2%和99.5%,平均多态信息含量分别为0.320和0.366,平均标记指数分别为7.38和7.15。SRAP检测8个国家菊苣遗传多样性指数均高于ESTSSR检测结果;两种标记检测结果显著相关(r=0.729,P<0.05),结合聚类可将80份材料分为8个组群,来源相同国家的材料基本能聚在一起,但聚类结果与地理分布并不完全一致。说明供试菊苣存在丰富的遗传多样性,其中意大利菊苣种质遗传多样性最丰富;两种标记均能够有效鉴定菊苣种和苦苣种,均适用于菊苣属物种遗传多样性及亲缘关系的鉴定。SRAP标记具有较高的多态性位点检测能力和较高的标记效率。 To study genetic diversity and genetic relationship of eighty materials of Cichorium germplasm originated from 15 countries included China, Italy, France and the United States, etc. , two molecular markers of SRAP and EST - SSR were used to detect the samples. The results showed that 257 and 218 bands were amplified by SRAP and EST - SSR, respectively. The average percentages of polymorphic loci were 96.2%, 99.5%, and the average polymorphic information contents were 0. 320 and 0. 366, and the average marker indexes were 7.38 and 7.15, respectively. The genetic diversity indices of Cichorium intybus L. from 8 Countries that detected by using SRAP were higher than those that detected by using EST - SSR, and the detection results of two methods were significantly correlated ( r = 0. 729, P 〈 0.05 ). Eight materials could be divided into 8 groups combined with clustering analysis, and the materials in the same country could be clustered together, but the clustering results were not completely consistent with the geological distribution. The results indicate that there is abundant genetic diversity a- mong the tested materials of Cichorium intybus L. , in which the Cichorium germplasm from Italy has the richest genetic diversity among 8 countries. The SRAP and EST - SSR markers can effectively identify Cichorium intybus L. and Cichorium endivia L. , and are suitable for the identification of genetic diversity and genetic relationship of Cichorium species, but the SRAP marker has higher capability for detecting the polymorphic loci and higher marking efficiency compare with the EST - SSR.
