RNA-Sequencing and Its Application in Medicinal Plants

摘要 RNA-Seq是一种新崛起的转录组学研究方法,新一代测序技术(NGS)能够更快速、更准确地为人们提供更多的生物体转录信息,已被广泛应用于医学研究、生物学研究、药物研发和临床研究等领域。药用植物转录组是现今研究药用植物基因组最活跃的领域之一,我们可从整体水平了解细胞中基因表达情况及其调控规律。对转录组的研究将有助于解决遗传育种和基因进化等诸多方面的问题。本综述简要介绍了转录组测序的原理,并从功能基因挖掘、SSR分子标记开发和代谢途径探索等3个方面综述了国内外近5年的转录组测序在药用植物上的应用进展,为药用植物的研究提供了更多基础数据。 RNA-Sequencing (RNA-Seq) is a newly-developed method in transcriptome research, which can afford transcription information more quickly and more accurately by using Next-generation Sequencing (NGS) technology which has been widely used in the areas of medical research, biological study, drug development and clinical application. The transcriptome of medicinal plants is one of the most active fields in the research of medicinal plant genome, from which we can know the expression and regulation rules of cells at integral level. The study of transcriptome has great significance in solving the problem of genetic breeding, gene evolution and so on. In this review, the principle of RNA-sequencing was briefly introduced and the application of RNA-sequencing in medicinal plants in the past five years at home and abroad was reviewed from three aspects including mining of functional genes, development of SSR markers and exploration of metabolic pathways, which would provide more basic data for medicinal plant studies.
