摘要
本文用5种分离培养基从大连海域潮间带采集的2种海绵中分离培养放线菌,以副溶血弧菌、哈维弧菌和鳗弧菌3种海水养殖病原菌为指示菌对放线菌进行抑菌活性的筛选,并基于16S rDNA序列对筛选出的抑菌活性较强的海绵放线菌进行多样性分析。结果表明:从海绵中共分离出57株海绵放线菌,以寡营养的M2培养基分离效果最好。抑菌活性筛选有55株对3株指示菌中的至少1株具有抑菌活性;有11株抑菌活性较强,对3株指示菌均具有抑菌活性,初步鉴定均属于链霉菌属。由此表明,海绵放线菌多能够产生对海水养殖病原菌较强的抑菌活性,是天然活性物质的重要来源,在海水养殖应用方面具有较大的开发价值。
This text isolated actinomycetes from 2 kinds of sponge taken from Dalian ocean intertidal zone with 5 kinds of separation medium.The antibacterial activity of sponge-associated aetinobacteria was screened with Vibrio parahaemolyticus, Vibrio harveyi and Vibrio anguillarum as indicator bacteria.The diversity analysis of sponge-associated actinobacteria with strong antibacterial activity was conducted based on 16S rDNA sequence.The study showed that 57 strains in total were isolated from the sponges, and the separation effect of oligo-culture M2 was the best.The result of antibacterial activity screening found that 55 strains had antibacterial effect on at least one of the 3 indicator bacteria.There were 11 strains preliminarily identified as Streptomyces sp. having stronger antibacterial effect on all the 3 indicator bacteria.The results showed that most of the sponge-associated actinobacteria which was an important source of bioactive natural products had quite strong inhibition on mariculture pathogen.h has a great development value in the field of maricuhure.
