肿瘤特异性γ δ T细胞扩增体系的建立及其抗肿瘤活性研究 被引量:2

摘要 目的:建立一种体外诱导肿瘤细胞特异性γδ T细胞的培养体系。方法:从人外周血中分离单个核细胞(PBMCs),用不同浓度的磷酸抗原(IPP)与rhIL-2联合刺激γδ T细胞增殖,观察并收集细胞,采用流式细胞仪检测刺激后CD3^+Vγ9^+的γδ T细胞数量以及比例,并计算其扩增效率。结果:经15天扩增培养后,高浓度组10μg/ml的IPP联合500IU/ml rhIL-2刺激得到的γδ T细胞的扩增效率最高,且扩增培养至第9天的γδ T细胞对肿瘤细胞有一定的杀伤活性。结论:适宜浓度磷酸抗原(IPP)联合rhIL-2能有效体外诱导具有较强抗肿瘤活性γδ T细胞的扩增。 Objective To amply tumor cells specific peripheral blood γ δ T T from tumor patients. Methods: Isolatingperipheral monocytes (PBMCs) from peripheral blood and amplifying yh Tce l ls by using IPP plus rhIL -2. Analy-zing proportion and total cell amount of CD3 + V "γ δ + T cells in total PBMCs by using flow cytometry approach. Explo-ring the tumor cel ls killing activities of activated CD3 + V " γ δ+ T cells by analyzing the target cells viability using Cell Counting Kit - 8 . Results : After 15 days culturing, the group cultured with the medium supplemented with 10μg/ml IPP plus 500IU/ml rhIL - 2 showed highest amplifying ef f icacy.γ δ T ce l ls cultured in the 9 th day showed optimal tumor cells killing activities.γ δ T activated with cell lysates from tumor cells DU145 showed specific cell killing activ-ities. Conclusion : Tumor cells targeting γ δ T could be induced and amplified with optimized IPP plus rhIL - 2 formu-la , which could be a potential tool for immune ce ll therapy.
