秋茄内生细菌多样性及抑制甘蔗黑穗霉菌活性研究 被引量:15

摘要 目的系统地对广西北海金海湾红树植物秋茄(Kandelia candel)内生细菌的分布特征与物种多样性进行分析,开展细菌代谢粗提物对甘蔗黑穗霉菌的抑制活性研究,为开发新型生物类农药及生物膜制剂奠定基础。方法采用7种分离培养基,从秋茄各部位组织中分离可培养细菌;通过16S rRNA基因序列的系统发育学分析其物种多样性。采用牛津杯法研究内生细菌对甘蔗黑穗霉菌的抑制效果。阳性菌株通过PCR扩增筛选次级代谢产物生物合成基因PKS I、PKS II和NRPS。结果经形态学分析及16S rRNA基因序列对比后,从秋茄各组织部位中分离到50株内生细菌,隶属于21科27属。从秋茄叶子中分离到BGMRC2075菌株与已有细菌物种典型菌株的全长16S rRNA基因相似性为96.12%,提示其为潜在新种。37株细菌对甘蔗黑穗霉菌有抑制作用。其中,16株菌可能存在至少1种NRPS、PKS I和PKS II生物合成基因簇。结论秋茄中可培养细菌具有较高的物种多样性,蕴藏着潜在新物种资源,且富含抑菌活性的菌株。 Objective To investigate the distribution and the diversity of endophytic bacteria of Kandelia candel collected from Beihai city and study the antifungal activities of their fermentation products against Ustilago scitaminea Sydow. Methods The endophytic bacteria were isolated from Kandelia candel by using seven different isolation media. The 16S rRNA gene sequencing was employed to explore the diversity. The effects of the fermentation products from bacteria against Ustilago scitaminea Sydow were tested with the method of Oxford cup. Preliminary screening of PKS I, PKS II and NRPS genes were carried out by PCR and agarose gel electrophoresis. Results A total of 50 strains of endophytic bacteria were obtained from all tissues and they were classified into 27 genera and 21 families. The BGMRC 2075 isolated from the leaves was a potential new species. Among them, the crude extracts of the strains showed strong inhibition effect against Ustilago scitaminea Sydow. PCR amplication from 16 out of 37 positivestrains showed a band of the expected size for PKS I, PKS II or NRPS gene. Conclusion Endophytic bacteria of Kandelia candel are genetically diverse and most of them showed strong inhibitory effects against Ustilago scitaminea Sydow.
