摘要
目的:建立测定荆防止痒颗粒中升麻素苷和5-O-甲基维斯阿米醇苷含量的高效液相色谱方法。方法:以Phenomenex Synergi C18(250 mm×4.6 mm,4μm)为色谱柱,流动相采用乙腈(A)-水(B)梯度洗脱:0~15 min,15%A;15~35 min,15%~35%A,检测波长为254 nm,流速为1.0 m L·min-1,柱温为35℃,进样量为10μL。结果:升麻素苷的进样量在0.049 735 96~1.243 399μg范围内呈现良好的线性关系(r=0.999 9),平均回收率为98.43%,RSD=0.59%(n=6);5-O-甲基维斯阿米醇苷的进样量在0.048 52~1.213μg范围内呈现良好的线性关系(r=0.999 9),平均回收率为98.85%,RSD=1.30%(n=6)。结论:该方法专属性强,准确度高,可用于荆防止痒颗粒中升麻素苷和5-O-甲基维斯阿米醇苷的含量测定。
Objective: To establish a HPLC method for determination of prim-O-glucasylcimifugin and 5-O-methylvisammioside in Jingfang Zhiyang Granule. Methods: The determination was performed on a Phenomenex Synergi C18 column (250 mm ×4.6 mm,4 μm) with the mobile phase consisting of Acetonitrile (A) -water (B) with gradient elution. The detection wavelength was set at 254 nm. The flow rate was 1.0 mL · min^ -1. The column temperature was 35 ℃. The injection volume was 10 μL. Results: The linear range of prim-O-glucasylcimifugin and 5-O-methylvisammioside were 0.049 735 96 -1. 243 399 μg (r =0. 999 9) and 0. 048 52 - 1. 213 μg(r =0. 999 9). The average recovery were 98.43 % ( RSD = 0.59 % , n = 6 ) and 98.85 % ( RSD = 1.30% , n = 6 ). Conclusion: The standard can be used for quality control of Jingfang Zhiyang Granule.
