摘要
目的:探讨健脾补肺化痰方对幼龄哮喘大鼠肺组织中TGF-β_1、c-fos表达的影响。方法:将120只雄性SD幼龄大鼠,随机分为正常组4周(Z4)、8周(Z8)、12周(Z12),模型组4周(M4)、8周(M8)、12周(M12),健脾补肺化痰方组4周(J4)、8周(J8)、12周(J12),地塞米松组4周(D4)、8周(D8)、12周(D12),每组30只,采用卵白蛋白(OVA)雾化激发建立哮喘大鼠模型,用Masson染色法及Img-Pro6.0图像分析系统,分别测定上述时间段各组大鼠肺组织中WAt/Pbm、WAi/Pbm及WAm/Pbm等气道形态学参数,同时采用逆转录聚合酶链反应(RT-PCR)分别检测各组大鼠肺组织中TGF-β_1、c-fosm RNA的表达。结果:与正常组相比,模型组肺组织中TGF-β_1、c-fos的表达及WAt/Pbm、WAi/Pbm、Wam/Pbm等气道形态学参数均有不同程度升高(P<0.05);与模型组相比,各治疗组以上指标均有不同程度下调(P<0.05);健脾补肺化痰方组与地塞米松组比较,差异无显著性(P>0.05)。结论:健脾补肺化痰方可通过下调TGF-β_1、c-fos的表达实现对气道重塑的抑制。
Objective:To investigate the effect of Jianpi Bufei Huatan Decoction on expression in lung tissue of young asthmatic rats TGF-β1,c-fos. Methods:120 young male SD rats were randomly divided into normal group 4 weeks(Z4),8 weeks(Z8),12 weeks(Z12),model group 4 weeks(M4),8 weeks(M8),12 weeks(M12),Jianpi Bufei Huatan Decoction group 4 weeks(J4),8 weeks(J8),12 weeks(J12),dexamethasone group 4 weeks(D4),8 weeks(D8),12 weeks(D12),n=30,using ovalbumin(OVA)atomization inspired rat model of asthma,with Masson staining and image analysis system Img-Pro 6.0 were measured above-mentioned period the lung tissue of rats WAt/Pbm,WAi/Pbm and WAm/Pbm and other airway morphological parameters,while using reverse transcription polymerase chain reaction(RT-PCR)expression in the lung tissue of rats with TGF-β1,c-fosm RNA were detected. Results:Compared with the normal group,model group,the TGF-β1 in lung tissue,c-fos expression and WAt/Pbm,WAi/Pbm,Wam/Pbm and other airway morphological parameters varying degrees increased(P〈0.05);compared with the model group,the treatment group above indexes varying degrees reduced(P〈0.05);Jianpi Bufei HuatanDecoction group and dexamethasone group,the difference was not significant(P〉0.05). Conclusion:Jianpi Bufei Huatan Decoction through down-regulating TGF-β1,the expression of c-fos to achieve inhibition of airway remodeling.
