摘要
利用Design-Expert软件,在前期单因素试验的基础上,采用Plackett-Burman设计和中心组合设计对双孢蘑菇新菌株W38液体菌种培养基及培养条件进行优化。根据响应面分析法建立模型并确定其最佳的培养基和培养条件为:小米粉49.90g·L^(-1),黄豆粉13.60g·L^(-1),KH_2PO_4 2.00g·L^(-1),MgSO_4·7H_2O 1.067 5g·L^(-1),初始pH 6.5,发酵时间7d,发酵温度24℃,摇瓶转速180r·min^(-1)。在此条件下,双孢蘑菇菌株W38液体菌种的菌丝体每100mL生物量可达2.35g,与初始培养条件下的菌丝体生物量相比提高了3.05倍,平均菌丝生长速度加快了14.7%。
Based on the results obtained by aprevious single factor test,medium formulation and conditions for the fermentation of Agaricus bisporus W38 were optimized by the Plackett-Burman experimental design and the central composite design with Design-Expert software.A regression equation was obtained using the response surface methodology.The optimized medium was formulated toconsistof 39.90g·L^(-1),13.60g·L^(-1),K_2HPO_4 2.0g·L^(-1),and MgSO_4·7H_2O 1.067 5g·L^(-1).The fermentation was designed to be carried out with an initial pH of 6.5and maintained at 24℃ with a constant shaking in flask at 180r·min^(-1) for 7d.A mycelial biomass 100 mL of 2.35 g,which was 3.05-foldof the original design,and an average mycelial growth rate improvement by 14.7% were obtained.
