摘要
研究香菇多糖中半乳糖醛酸的含量测定和抗氧化作用。采用硫酸-咔唑比色法与间羟基联苯法测定半乳糖醛酸的含量,采用羟基自由基、总还原力、1,1-二苯基苦基苯肼(DPPH)自由基、超氧阴离子自由基、2,2'-联氮双(3-乙基苯并噻唑啉-6-磺酸)二铵盐(ABTS)自由基的清除作用考察5种不同体系香菇多糖的抗氧化活性。标准半乳糖醛酸质量浓度在26.15~78.45μg/m L之间呈良好的线性关系,r=0.999 4,平均加样回收率为99.87%,RSD为0.11%,最低检出限为4.086μg/m L,糖醛酸含量为30.25μg/mg;香菇多糖质量浓度在0.2~1.0 mg/m L范围内抗氧化作用大小依次为羟基自由基、总还原力、DPPH,超氧阴离子和ABTS没在IC50范围内。硫酸-咔唑比色法简便易行、准确性高、重现性好,且多糖5种体系验证香菇多糖具有一定的抗氧化能力。
Lentinan has anti-tumor, anti-virus, improving the immune function and stimulating the formation of interferon and other functions The present study was to determinate the content of lentinanuronic acid and to examine the antioxidation activity of Lentinan. The content of uronic acids was determined according to the method of CarbazolesulfiLric acid colorimetry and m-hydroxydiphenyl. The antioxidant effect of Lentinan was detected by scavenging hydroxyl radical, reducing power assay, DPPH radicals, superoxide anion free radical O2-, and ABTS radical method with Vc as a positive control. The results showed that a good linearity relation was existed between the absorbency and the concentration of standard galacturonic acid in the range of 26.15-78.45 μg/mL (r = 0.999 4), and the average recovery was 99.87%, RSD was 0.11%, and the lowest detection limit was 4.086 μg/mL. The content of uronie acids in Lentinan was 30.25 p,g/mg. The concentration of Lentinan in the range of 0.2-1.0 mg/mL had a certain antioxidant effect, which was in the order of scavenging hydroxyl radical, reducing power assay and DPPH radicals. The superoxide anion free radical and ABTS were not within the scope of IC50. It was concluded that the method of Carbazolesulfuric acid colorimetry and m-hydroxydiphenyl to determinate the lentinanuronic acid content in Lentinan was simple, available, accurate and reproducible, and the Lentinan had been proved to have certain antioxidation effect by 5 kinds of method.
