复方丹益莲抗家兔动脉粥样硬化的作用及机制

摘要 目的研究复方中药丹益莲防治动脉粥样硬化的作用及机制。方法采用高脂饲料喂养及免疫损伤方法建立家兔动脉粥样硬化模型,随机分为模型组、丹益莲高剂量组、低剂量组、阳性药物辛伐他汀组,另设普通饲料喂养的家兔为正常对照组。药物干预8周后,心脏穿刺取血测定血浆总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)和载脂蛋白b(Apo-b)。处死家兔,取冠状动脉HE染色观察粥样硬化斑块的形成;取肝脏测定谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、丙二醛(MDA)及超氧化物歧化酶(SOD);采用Western blot方法检测主动脉组织中血管细胞黏附分子1(VCAM-1)、基质金属蛋白酶2(MMP-2)及肿瘤坏死因子α(TNF-α)的表达水平。结果与模型组比较,丹益莲给药组血管内膜斑块明显减小,血浆TG、LDL及Apo-b水平显著降低;肝脏GSH、GSH-Px、CAT和SOD含量明显升高,MDA含量显著降低;主动脉组织中VCAM-1、MMP-2和TNF-α表达水平下降。结论复方丹益莲能显著抑制动脉粥样硬化斑块的形成,具有明显的抗动脉粥样硬化作用,其机制可能与降低血脂、减轻氧化应激及抗炎等有关。 Objective To investigate the effects of compound Danyilian on atherosclerosis in rabbits. Methods The atherosclerosis in rabbits was established with high fat diet and immune injury. The rabbits were randomly divided into model group, Danyilian high dosage and low dosage groups, positive control simvastatin group. Rabbits fed with normal diet were used as normal control. After 8 weeks of drug administration, blood lipid levels including the levels of total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL) and apolipoprotein b (Apo-b) in plasma, and the levels of glutathione (GSH), glutathione peroxidase (GSH-Px), catalase (CAT), malondialdehyde (MDA) and superoxide dismutase (SOD) in liver were determined. HE staining of coronary artery was performed to observe the aortic plaque formation. The expression of VCAM-1, MMP-2 and TNF-ct in aorta tissues was detected by Western blot. Results Compared with the model group, the plaque formation in the coronary artery of the Danyilian treated rabbits was highly inhibited; levels of TG, LDL and Apo-b in plasma decreased obviously, and levels of GSH, GSH- Px, CAT and SOD in liver increased markedly, while the amount of MDA in liver decreased. In addition, expression of VCAM-1, MMP-2 and TNF-a in aorta tissues was reduced. Conclusion Danyilian showed obvious anti-atherosclerotic effect through the inhibition of aortie plaque formation, and the protective effects might be related to the decrease of blood lipid, improvement of oxidative stress as well as anti-inflammation.
