摘要
目的探讨肺癌组织中SDF-1及CXCR4蛋白表达与肺癌生物学特性的关系。方法采用免疫组化方法检测80例肺癌组织及40例癌旁肺组织中SDF-1及CXCR4蛋白的表达,并用Western blot法对两组组织中SDF-1及CXCR4的蛋白表达水平进行验证。分析SDF-1及CXCR4表达情况与肺癌生物学特性的关系。结果免疫组化结果显示,相比癌旁肺组织,SDF-1和CXCR4蛋白在肺癌组织中的表达水平升高,差异有统计学意义(P<0.05)。Western blot检测结果显示,肺癌组织中SDF-1和CXCR4蛋白表达阳性率高于癌旁组织,差异有统计学意义(P<0.05);肺癌组织中SDF-1蛋白表达与CXCR4蛋白表达呈正相关(r=0.501,P=0.026)。沉默SDF-1与CXCR4表达,肺癌细胞凋亡率增加。结论肺癌组织中SDF-1及CXCR4蛋白均呈高表达,且二者表达呈正相关,在肺癌的发生、发展中可能存在协同作用。
Objective To investigate the expression of SDF-1 and CXCR4 protein in lung cancer tissues, and their relationship with biological characteristics. Methods The expression of SDF-1 and CXCR4 protein were examined by immunohistochemical methods re- spectively in 80 cases of lung cancer and 40 cases of adjacent lung cancer tissues. The expression levels of SDF-1 and CXCR4 protein were verified by Western blot respectively in the two groups. The relationship of the SDF-1 and CXCR4 protein expression with the biological property of lung cancer was analyzed. Results The expression levels of SDF-1 and CXCR4 protein were different in lung cancer and ad- jacent lung cancer tissues (P 〈 0.05). The SDF-1 and CXCR4 protein expression were significantly higher in lung cancer tissues than in the adjacent lung cancer tissues (P 〈 0.05). The expression of SDF-1 was positively correlated with that of CXCR4 protein in lung cancer (r = 0.501, P = 0.026). The apoptosis rate of lung cancer cell was increased after SDF-1 and CXCR4 expression were silenced. Conclusion SDF-1 and CXCR4 proteins were highly expressed in lung cancer tissues. The expression of SDF-1 and CXCR4 were positive associated, and they may play synergistic effect in the occurrence and development of lung cancer.
