摘要
目的探讨^(12)C^(6+)重离子射线对人肝癌HepG2细胞迁移、侵袭和黏附能力的影响及相关机制。方法选择对数生长期HepG2细胞,经0、1、2、4 Gy剂量^(12)C^(6+)重离子射线辐照后,用Transwell法检测HepG2细胞迁移能力和侵袭能力,MTS法(内盐法)测定黏附能力,蛋白免疫印迹法检测基质金属蛋白酶-2(MMP-2)、MMP-9蛋白表达水平。结果 ^(12)C^(6+)重离子射线辐照后,与0Gy组相比,随着辐照剂量的增加,HepG2细胞迁移能力显著下降,侵袭能力也随之下降(P<0.05);HepG2细胞黏附能力增强(P<0.05);HepG2细胞中MMP-2和MMP-9表达量下调(P<0.05)。结论 ^(12)C^(6+)重离子辐照能增强HepG2细胞的黏附能力,抑制HepG2细胞的迁移能力和侵袭能力,这种作用可能与其下调MMP-2和MMP-9的表达有关。
Objective To investigate the effects of ^12C^6+ heavy ion irradiation on migration, invasion and adhe- sion of human hepatocellular carcinoma HepG2 cells and its related mechanisms. Methods The logarithmic HepG2 cells were irradiated by 0, 1, 2, 4 Gy of ^12C^6+ heavy ion ray and then the migration and invasion were determined by Transwell method, the adhesion ability was detected by MTS method (inner salt method), and the matrix metalloproteinases-2 (MMP-2) and MMP-9 protein expression was detected by Western blot. Re- sults After ^12C^6+ heavy ion rays irradiation, compared with the 0 Gy group, with the increase of radiation dose, HepG2 cell migration ability decreased significantly, the invasion ability also decreased (P 〈 0.05); HepG2 cell adhesion ability enhanced (P 〈 0.05); the amount of MMP-2 and MMP-9 expression in HepG2 cells were down-regulated (P 〈 0.05). Conelusion ^12C^6+ heavy ion irradiation can enhance the adhesion ability and inhibit the migration and invasion ability of HepG2 cells, which may be related to the down-regulation of MMP-2 and MMP-9 expression.
