聚维酮联用轻质碳酸钙用于槲皮素固体分散体的制备及评价 被引量:6

Preparation and Evaluation of Quercetin Solid Dispersion by Combining Povidone with Precipitated Calcium Carbonate

摘要 目的:为提高槲皮素的稳定性及溶出度,以聚乙烯吡咯烷酮(PVP)联用轻质碳酸钙(PCC)为载体制备槲皮素固体分散体。方法:采用溶剂法制备固体分散体,以电子扫描显微镜法、差示扫描量热法对槲皮素固体分散体微观结构、物理特征等进行表征分析,利用小杯法考察槲皮素固体分散体的体外溶出行为,通过稳定性影响因素试验考察其稳定性。结果:固体分散体中槲皮素在载体中以无定形状态分散;在15 min时,槲皮素固体分散体[药质比1∶3,PVP-PCC(2∶1)]的药物累积释放率>90%;稳定性影响因素试验表明固体分散体中药物不受强光、高湿、高温条件的影响。结论:PVP联用PCC作为混合载体所制备的槲皮素固体分散体具有速释效果,能显著改善槲皮素的溶出行为,且稳定性良好。 Objective: In order to improve the stability and solubility of quercetin,quercetin solid dispersion was prepared with the combination of polyvinyl pyrrolidone(PVP) and precipitated calcium carbonate(PCC) as carriers. Method: The quercetin solid dispersion was prepared by solvent evaporation method. The microscopic structure and physicochemical properties of the solid dispersion were analyzed by scanning electron microscopy(SEM) and differential scanning calorimetry(DSC),in vitro dissolution behavior and stability of the solid dispersion were also investigated by small cup method and stability test,respectively. Result: Quercetin in the solid dispersion was amorphous form in the carriers. The cumulative release rate of quercetin in the solid dispersion [drug-matrix(1 ∶ 3), PVP-PCC(2 ∶ 1) ] was 〉90% at 15 min. The stability of quercetin solid dispersion was not affected by light, high humidity and high temperature. Conclusion: Quercetin solid dispersion,prepared by combining PVP with PCC as a hybrid carrier,has an immediate release effect; and it can significantly increase the dissolution behavior of quercetin with good stability.
