葛根素对前脂细胞分化及胰岛素抵抗模型糖摄取的影响 被引量:3

摘要 目的探讨葛根素对3T3-L1前脂细胞分化及胰岛素抵抗模型糖摄取的影响。方法 (1)实验分为葛根素3,10,30,100,300μmol/L组、空白组、罗格列酮20μmol/L组,各加药组分别给予相应浓度药物对3T3-L1前脂细胞进行干预,空白组常规培养,采用油红O染色法检测各组3T3-L1前脂细胞分化情况。(2)实验分为葛根素3,10,30,100,300μmol/L组、空白组、罗格列酮20μmol/L组,除空白组外,其余各组应用地塞米松对3T3-L1前脂细胞诱导进行胰岛素抵抗模型的建立,然后分别给予相应浓度药物对3T3-L1前脂细胞进行干预,空白组常规培养,检测各组细胞葡萄糖摄取量。结果葛根素3,10μmol/L组细胞分化成脂的相对变化率与空白组比较差异均无统计学意义(P均>0.05),葛根素30,100,300μmol/L组和罗格列酮20μmol/L组细胞分化成脂的相对变化率均明显高于空白组(P均<0.05);葛根素3,10μmol/L组胰岛素抵抗模型3T3-L1前脂细胞葡萄糖摄取相对变化率与空白组比较差异均无统计学意义(P均>0.05),葛根素30,100,300μmol/L组和罗格列酮20μmol/L组胰岛素抵抗模型3T3-L1前脂细胞葡萄糖摄取相对变化率均明显高于空白组(P均<0.05)。结论低浓度葛根素对3T3-L1前脂细胞影响较小,高浓度的葛根素能够促进3T3-L1前脂细胞分化及提高胰岛素抵抗模型3T3-L1前脂细胞葡萄糖摄取率。 Objective It is to investigate the effects of Puerarin on differentiation of preadipocytes 3T3-L1 and glucose up-take of these cells with insulin resistance. Methods The preadipocytes 3T3-L1 were divided into Puerarin groups (3, 10, 30, 100, 300 jxmol/L) , blank group, 20 jxmol/L Rosiglitazone group, the preadipocytes 3T3-L1 were treated in every treatment group with respective medicine, the cells in blank group were cultured normally. The differentiation of preadipocytes 3T3-L1 was detected by oil red 0 staining. The preadipocytes 3T3-L1 were divided into Puerarin groups (3, 10, 30, 100, 300 μmol/ L),blank group, 20 ( xmol/L Rosiglitazone group, all the cells in every group except blank group were given Dexamethasonum to establish insulin resistance models which were treated with respective medicine, the cells in blank group were cultured normally, glucose uptake of the cells was detected. Results There was no difference in relative change rates of preadipocytes in 3 , 10 μmol/L groups compared with blank group(P 〉0. 05) , and the rates in 30, 100, 300μmol/L Puerarin groups and 20 ( xmol/L Rosiglitazone group were higher than that in blank group (P 〉0. 05). There was no difference in relative change rates of glucose uptake of insulin resistance models of 3T3-L1 in 3,10 μmol/L groups compared with blank group(P 〉0. 05) , and the rates in 30, 100, 300 ( xmol/L Puerarin groups and 20 μmol/L Rosiglitazone group were higher than that in blank group (P 〉 0. 05 ) . Conclusion Low concentration of Puerarin has less effect on 3T3-L1 , high concentrations of Puerarin can promote dif-ferentiation of 3T3-L1 and improve the glucose uptake rate in insulin resistance models of 3T3-L1.
