Frizzled 3、Frizzled 4基因对黑线仓鼠A:CHA白化性状产生影响的初步分析

Analysis of the function of Frizzled 3、Frizzled 4 genes in the albino phenotype of Cricetulusgriseus

摘要 为了比较Wnt通路重要结合受体Frizzled家族的Frizzled 3、Frizzled 4受体蛋白基因在黑线仓鼠及其白化突变系(A:CHA)皮肤组织的表达差异,分析这两种基因对白化性状产生的影响,试验提取黑线仓鼠和A:CHA的皮肤组织总RNA,利用普通PCR方法筛选引物,通过实时荧光定量PCR技术比较黑线仓鼠和A:CHA皮肤组织Frizzled 3、Frizzled 4基因表达量的差异。结果表明:黑线仓鼠皮肤中的Frizzled 3、Frizzled 4基因mRNA的相对表达量分别是A:CHA组织中的4倍、5倍。Frizzled 3、Frizzled 4基因在A:CHA皮肤组织中的表达量下调。说明Frizzled 3、Frizzled 4基因与A:CHA白化性状的产生存在一定关联。 In the current study, investigation and analysis of the expression level of Frizzled 3, Frizzled 4 of animportantreceptor of Wnt protein between Cricetulusgriseus and A : CHA and the funetionon the albino phenotype were carried out. The total mRNA of skin tissue in the Cricetulusgriseus and A : CHA was extracted, and the expression levels of Frizzled 3 and Frizzled 4 were analyzed by using real time PCR. The results showed that the ratio of expression level of Frizzled 3, Frizzled 4 between Cricetulusgriseus and A : CHA were revealed to be increased by 4 and 5 times. These data demonstrated that the gene expression level of Frizzled 3, Frizzled 4 in the A:CHA were lower than that in the Cricetulusgri- seus. The resulting data suggests the notion the two genes may be contribute to theAlbino phenotype of A:CHA.
