摘要
目的:检测消痰和胃方对胃癌前病变大鼠胃黏膜组织NF-κB通路的影响,探讨该方干预胃癌前病变的机制。方法:清洁级Wistar雄性大鼠,空白对照组和模型组,模型组采用以化学干预为主的多因素造模方法,共32周。模型组再分为模型对照组、维酶素组、消痰和胃方组。干预6周后采用Western blot检测大鼠胃黏膜组织中p-NF-κB p65蛋白表达的差异。结果:与模型对照组相比,消痰和胃方组p-NF-κB p65表达降低(P<0.05),维酶素组无统计学意义(P>0.05),并且消痰和胃方能改善胃癌前病变大鼠整体情况,较维酶素更优。结论:消痰和胃方防治胃癌前病变的机制有可能通过干预NF-κB通路实现的,值得进一步深入研究。
Objective : To investigate the impact of Xiaotan Hewei Prescription on the effect of NF - KB pathway in precancerous lesions of gastric carcinoma rats and explore the potential mechanisms. Methods: Male Wistar rats of clean grade were randomly divided into normal control group and model group. The MNNG was used to establish the model of precancerous lesions of gastric cancer in rats indueed by multiple factors ahout 32 weeks. The successlul model male Wistar rats were divided into three groups randomly:the model control group, the Vitaeoenzyme group, the Xiaotan Hewei Prescription group. We gave the medicine to each group 6 weeks. Western blot was used to observe the expression of p - NF - KB P65 of gastric mucosa. Results : The expression of p - NF - KB P65 protein had statistically significant difference between the Chinese herb group and the other two groups (the model control group, the Vitaeoenzyme group) (P 〈 0. 05 ) and there was no distinct difference between the Vitacoenzyme group and the model control group(P 〉0. 05). The effect of Xiaotan Hewei Prescription was much better than the Vitaeoenzyme. Conclusion: Xiaotan Hewei Prescription could reduce the p - NF - KB P65 expression of NF - KB signal pathway and block the abnormal activation of it. The results illustrated that Xiaotan Hewei Prescription could reverse PLGC which might be achieved through influenee of the NF - KB signal pathway.
