摘要
目的筛查人大肠癌肿瘤组织与相应癌旁正常组织差异表达基因。方法应用NimbleGen基因芯片检测4例大肠癌肿瘤组织及相应癌旁正常组织基因表达谱,并以RT-PCR技术对部分基因芯片检测结果进行验证,利用生物信息学方法对检测结果进行分析。结果大肠癌肿瘤组织和癌旁正常组织差异表达基因共5 042条,其中表达水平上调3 399条,下调1 643条,这些基因涉及多个信号通路。结论本研究结果可为寻找大肠癌分子标记物和探索大肠癌发生的分子机制提供实验依据。
Objective To screen the differentially expressed genes in human colorectal cancer(CRC) tissue. Methods Nimble Gen gene expression array information were used to detect the gene expressions of CRC tissue paired with normal colorectal tissue. The microarray findings were confirmed by real-time quantitative reverse transcriptase-polymerase chain reaction(RT-PCR).The results were analyzed by bioinformatic technique. Results Compared with normal tissues, a total of5042 genes with more than 2-fold differential expression were detected in CRC tissues, of which 3399 genes were up-regulated and1643 genes were down-regulated. These genes involved multiple signaling pathways. Conclusion The results may provide experimental basis to seek for molecular markers and to investigate the molecular mechanism of colorectal cancer.
