摘要 目的研究Fibulin-5对肺癌发生发展的作用及其机制。方法将Fibulin-5过表达质粒和空白质粒转入肺癌细胞中,Western blot和免疫荧光检测β-catenin在核内的定位;在低表达Fibulin-5的肺癌细胞系中过表达Fibulin-5,同时在高表达Fibulin-5的肺癌细胞系中敲低Fibulin-5,Western blot检测p-ERK、p-GSKβ和β-catenin及其下游蛋白的表达;在肺癌细胞中过表达Fibulin-5,进行裸鼠成瘤实验,观察Fibulin-5对肺癌细胞成瘤能力的影响,免疫组化检测两组肿瘤中p-ERK、p-GSKβ和β-catenin及其下游蛋白的表达;免疫组化检测肺癌临床样本中Fibulin-5和β-catenin的表达情况,并进行相关性分析。结果在肺癌细胞中过表达Fibulin-5后β-catenin的核内定位减少;Fibulin-5可以下调p-ERK/p-GSKβ/β-catenin信号通路;Fibulin-5抑制肺癌细胞成瘤能力;肺癌样品中Fibulin-5和β-catenin的表达成负相关。结论 Fibulin-5通过下调β-catenin抑制肺癌的发生发展。 Objective To study the effect and mechanism of Fibulin-5 on the occurrence and development of lung cancer. Methods Fibulin-5 expression plasmid and blank plasmid were transfected into lung cancer cells, Western blot and immunofluores-cence were used to detect the expression of beta -catenin in the nucleus; Over-express Fibulin-5 in lung cancer cell lines with low ex- pression of Fibulin-5, and knockdown Fibulin-5 in lung cancer cell lines with high expression of Fihulin-5, Western blot were used to detect the expression of p-ERK, p-GSK beta and beta -catenin and its downstream proteins; Over-express Fibulin-5 in lung cancer ceils to observe the effect of Fibulin-5 in lung cancer cell tumorigenicity, and use immunohistochemistry to detect the expression of p-ERK and p-GSK beta and beta -catenin and its downstream proteins of the two groups of tumors; immunohistochemistry was used to detect the expression of Fibulin-5 and beta-caten in clinical lung cancer samples, and analyse the relationship. Results Fibulin-5 reduced the nuclear localization of beta-catenin in lung cancer cells; Fibulin-5 can downregulate the p-ERK/p-GSK beta / beta-catenin signal- ing pathway; Fibulin-5 inhibits tumorigenicity of lung cancer cells; the expression of Fibulin-5 and beta -catenin in lung cancer sam- ples was negatively correlated. Conclusion Fibulin-5 inhibits the development of lung cancer by down regulating beta -eatenin.
