摘要
根据虎杖(Polygonum cuspidatum)中主要活性成分虎杖苷和白黎芦醇提取过程中效率低、耗时长等问题研究了匀浆提取虎杖苷和白藜芦醇工艺。以虎杖苷和白藜芦醇的提取总得率为指标,在单因素试验的基础上,利用响应面法设计了以乙醇体积分数、匀浆时间和液料比为影响因素的试验,获得了匀浆提取虎杖苷和白藜芦醇的优化工艺为提取溶剂64%乙醇溶液,匀浆时间5min,液料比14∶1(mL∶g),在此条件下虎杖苷和白藜芦醇的一次提取总得率为1.830%。与超声辅助提取和浸提法相比,匀浆提取具有绿色高效、速度快、能耗低等优点,在虎杖等中药活性成分的大规模提取方面具有应用前景。
As a traditional herbal medicine in China, tigen stick (Polygonum cuspidatum) has the effects of damp-inhibiting and wind-dispersing relieving, relieving cough and reducing sputum. Polydatin and resveratrol are the main bioactive constituents in P. cuspidatum. Polydatin is the precursor compound of resveratrol, and it can promote weight loss, provoke appetite in low-income mothers with young kids and enhance heart function and microcirculation, etc. Resveratrol is widely used in health-care products, cosmetics and food additives due to its antioxidant capacity, anti- inflammatory, anti-cancer and other various pharmaceutical properties. However, the extraction efficiency of resvera- trol and polydatin are relatively low, so the study on the rapid and efficient method of extraction is necessary. Homog- enate can improve pulverization efficiency because of powerful mechanical forces and liquid shearing forces. In addi- tion, it can also homogenate with solvent thoroughly and promote solvent disperse into the inner part of solid phase easily. Therefore, the homogenate technique was used to extract polydatin and resveratrol from P. cuspidatum in this study. Total extraction yield of polydatin and resveratrol was used as indicator. On the basis of singer factor experiment, the influence of ethanol volume fraction, extracting time and liquid-solid ratio on the total yield of polydatin and resveratrol were investigated by using Box-Behnken central composite design. The optimized extracting conditions were as follows : extracting solvent of 64% ethanol solution, extracting time of 5 min, liquid-solid ratio of 14 : 1 ( mL : g). Under the optimized conditions, the measured yield of targets was 1.83%. Compared to the ultrasonic assisted ex- traction and soaking extraction method, homogenate technique could have prospects for industrial scale-up applications in active ingredients extraction from P. cuspidatum and other Chinese herbal medicine, with the advantages of potential eco-friendly, higher efficiency and low energy consumption and so on.
