摘要 目的探讨前列腺素E1(PGE1)后处理对大鼠移植肝缺血再灌注损伤(IRI)的保护作用及其机制。方法建立SD大鼠肝移植模型,随机分为3组:IRI组、PGE1后处理组(PGE1组)及假手术组(S组)。检测各组血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、超氧化物岐化酶(SOD)及丙二醛(MDA)的含量;采用逆转录聚合酶链式反应(RT-PCR)方法检测移植肝组织内肿瘤坏死因子(TNF-α)信使核糖核酸(mRNA)的表达水平;苏木精-伊红染色(HE)观察移植肝病理形态学变化。结果供肝再灌注后2及6 h,PGE1组的大鼠血清ALT、AST和MDA水平及TNF-αmRNA的表达低于IRI组(P<0.05),血清SOD水平高于IRI组(P<0.05),光镜下PGE1组肝组织损伤较IRI组轻。结论 PGE1后处理可通过提高肝组织的抗氧化能力,降低TNF-α来减轻大鼠移植肝IRI。 Objective To study the protective effects and mechanisms of prostaglandin E1 (PGE1) post- conditioning in liver graft ischemia-reperfusion injury. Methods The liver transplantation rat model was used as ischemia-reperfusion injury animal model. The rats were randomly divided into three groups: ischemie reperfusion injury group (IRI), postconditional PGE1 treated group (PGE1) and sham operated group (S). The liver functional tests, SOD, MDA, TNF-α expression level and the histology were checked at 2 h and 6 h after reperfusion. Results The ALT, AST, MDA in serum and TNF-α expression level in liver were significantly lower in PGE1 group than in IRI group (P〈 0.05), the SOD was higher in the PGE1 group (P〈 0.05). The tissue damage was significantly decreased in PGE1 group. Conclusions Prostaglandin E1 postconditioning has significant protective effect in ischemia-reperfusion injury, and part of those benefit effects dependent on decreased TNF-α expression level and increased SOD production.
