摘要
目的:研究不同浓度苦参碱对生物膜状态下的白色念珠菌ALS基因mRNA表达量的影响。方法:采用白色念珠菌标准菌株ATCC76615为研究对象,经载玻片法形成白色念珠菌生物膜。在经不同浓度苦参碱干预下,通过RT-PCR法检测生物膜状态白色念珠菌ALS1/ALS4基因mRNA表达量的变化。结果:不同浓度苦参碱干预下生物膜状态白色念珠菌黏附相关基因ALS1/ALS4 mRNA表达减弱。1~8 mg/m L苦参碱干预下ALS1/ALS4的mRNA表达相对量分别为0.96~0.34/0.93~0.28。结论:苦参碱可抑制白色念珠菌粘附相关基因ALS1/ALS4 mRNA水平的表达。
Objective:To investigate the influence of different concentrations of matrine on the ALS gene expression in C. albicans biofilms. Methods:C. albicans collection strain ATCC76615 was obtained and biofilms were founded by slide method after following brief exposure to different concentrations of matrine in vitro. The levels of ALS1 and ALS4 mRNA expression were assessed by RT-PCR. Results : Matrine-treated ceils expressed lower levels of ALS1 and ALS4 mRNA than the cells grown in the absence of matrine. High ALS mRNA was observed in normal mature biofilm. When the cells were treated with matrine,ALS decreased. The relative ALS1 was 0. 96 and 0. 34 at matrine concentrations of 1 mg/mL and 8 mg/mL,respectively. While the relative ALS4 was 0. 93 and 0. 28 at matrine concentrations of 1 mg/mL and 8 mg/mL, respectively. Conclusion:Matrine could inhibit C. albicans biofilms expressed lower levels of ALS1 and ALS4 mRNA in vitro.
