摘要
目的:探讨熟地黄多糖对荷瘤小鼠肿瘤组织细胞色素C(Cytochrome C,Cyt-C)及天冬氨酸特异性的半胱氨酸蛋白水解酶(cysteinyl aspartate specific proteinases,Caspase-3)基因表达的影响。方法:采用Real-time PCR法检测肿瘤组织中Cyt-C m RNA及Caspase-3 m RNA的表达情况,采用Western blot法检测Cyt-C及Caspase-3蛋白质的表达情况。结果:熟地黄多糖低剂量组、中剂量组、高剂量组、阳性对照组和联合用药组肿瘤组织中Cyt-C和Caspase-3 m RNA的表达量分别是模型对照组的0.46、0.76;4.82、1.26;0.43、0.60;13.91、4.65;20.17、9.23倍。Cyt-C和Caspase-3蛋白质的表达量分别是模型对照组的0.31、0.90;1.82、1.46;0.62、0.87;2.60、1.54;2.80、1.60倍。结论:熟地黄多糖能促进肿瘤组织中Cyt-C和Caspase-3基因的表达。
Objective : To study the effect of radix rehmanniae polysaccharide on the expression of Cytochrome c (Cyt- C ) and cysteinyl aspartate specific proteinases (Caspase -3 ) genes in tumor- bearing mice. Meth- ods : Real Time - PCR was used to analyze the expression of Cyt - C and Caspase - 3 mRNA in tumor tissue. Western blot was used to analyze the expression of Cyt - C and Caspase - 3 proteins in tumor tissue. Results : The mRNA expression of Cyt - C and Caspase - 3 in the mice treated by low -, mid - , high - dose Radix re- hmanniae polysaccharide, positive control and drug combination was 0.46, 0.76 ; 4.82, 1.26 ; 0.43, 0.60 ; 13.91,4.65 ; 20.17, 9.23 times respectively than that of control mice. The protein expression of Cyt - C and Caspase - 3 in these five groups was 0.31,0.90 ; 1.82, 1.46 ; 0.62, 0.87 ; 2.60, 1.54 ; 2.80, 1.60 times higher than that of control mice. Conclusion:Radix rehmanniae polysaccharide can promote the expression of Cyt - C and Caspase - 3 mRNA and proteins in tumor tissues.
