摘要
目的研究灯盏花素(Breviscapine,Bre)对大鼠肺缺血再灌注损伤后细胞凋亡以及凋亡相关基因、蛋白表达的影响,探讨其可能的作用机制。方法通过夹闭左肺门45 min的方法,建立大鼠肺缺血再灌注损伤模型,术前30 min,分别给予不同浓度灯盏花素(12.5、25、50 mg/kg)和舒血宁(4 mg/kg),并设假手术组和模型组。再灌注2 h后,测定肺组织湿/干重比(W/D),TUNEL法检测肺细胞凋亡,RT-PCR法检测肺组织中bcl-2 mRNA、Bax mRNA表达,比色法测定肺组织中抗氧化酶活性和丙二醛(MDA)含量,Western blot法测定肺组织中NF-κB蛋白表达。结果与模型组比较,经灯盏花素干预能够显著降低肺缺血再灌注损伤大鼠肺组织W/D,减轻肺水肿,降低肺细胞凋亡指数(AI),改善肺细胞凋亡状况,上调bcl-2 mRNA表达,下调Bax mRNA表达,提高bcl-2/Bax比值,改善抗氧化酶(SOD、CAT)活性,降低MDA含量,下调NF-κB蛋白表达,上述作用均具有一定剂量依赖性。结论灯盏花素可能通过调节凋亡相关基因、蛋白表达而对肺缺血再灌注损伤大鼠肺细胞凋亡起到抑制作用,对肺缺血再灌注损伤起到一定的保护作用。
Objective To investigate the effects of breviscapine on lung cell apoptosis, the expression of apoptosis-related genes and protein in the rats with pulmonary ischemia-reperfusion injury, and to explore its possible mechanism. Methods The pulmonary ischemia-reperfusion model rats were made by clipping left lung door for 45 rain and were treated with Bre ( 12. 5,25,50 mg/kg) and Shuxuening (4 mg/kg) at 30 rain before the operation, and the sham operation group and model group were set up. Two hours after reperfusion, the wet/dry (W/D) ratio of lung tissue was examined ; the apoptosis of lung tissue cells were observed by TUNEL staining ; the expression of bcl-2 mRNA and Bax mRNA in lung tissue was determined;the activity of antioxidase and the content of MDA in lung tissue were deter- mined;the expression of NF-KB in lung tissue was determined by Western blot. Results Compared with model group, the lung tissue W/D of the rats in Bre-treated groups was significantly decreased, the AI was significantly decreased and the lung cells apoptosis was significantly improved;the expression of bcl-2 mRNA was significantly up-regulated and the Bax mRNA was significantly down-regulated;the ratio of bcl-2/Bax was significantly increased;the activity of SOD and CAT in lung tissue were significantly increased and the content of MDA was significantly decreased ; the expression of NF-KB protein was significantly decreased; all of the effects above-mentioned were some dose-dependent. Conclu- sion Bre has inhibitive effects on lung ceils apoptosis through altering the expression of apoptosis-related genes and pro- tein,which suggests that Bre has protective effects on the rats with pulmonary ischemia-reperfusion injury.
