摘要 建立了超高效液相-质谱联用(UPLC-MS/MS)法同时测定药用植物皂角中槲皮苷、槲皮素、山柰酚和芦丁含量的分析方法。优化的分析条件为:流动相:甲醇-水溶液(42∶58,V/V);流速:0.25 mL/min;柱温:25℃;进样量:10μL。采用电喷雾离子源进行电离,多离子反应监测(MRM)模式进行信号采集,采用外标法进行定量,结果表明,在不同检测范围内各目标化合物线性关系良好(R2>0.9997)。同时,应用建立的方法对比分析了4种目标化合物在皂角果荚和皂角刺两个部位的含量水平。 In this study, we established an UPLC-MS/MS method for the simultaneous determination of quercetin, quercetin, kaempferol and rutin in Gleditsia sinensis. By investigation of chromatography and mass spectrometry methods, the optimum analytical conditions: Mobile phase of methanol-water solution (42 : 58, V/V); flow rate, 0.25 mL/min; column temperature, 25 ℃ ; sample size, 10 μL. Using electrospray ionization (ESI) ionization, multiple reaction monitoring (MRM) mode for signal acquisition, quantified using the external standard method. The result showed that the test at different concentration range was a great linear relationship (R2〉 0.9997). Using this method, we assayed and tested the contents of the involved four compounds in thorn and pod, showing high sensitivity, good selectivity and shorter analysis time.
