摘要
仙人掌皂苷是仙人掌科植物中含有的一类重要生物活性成分。利用果胶酶辅助提取仙人掌总皂苷,以总皂苷提取率为评价指标,结合单因素实验和正交设计,探究pH值、酶添加量、酶解温度和酶解时间四个因素对总皂苷提取率的影响并进行验证实验。实验结果表明果胶酶辅助提取仙人掌总皂苷的最佳提取工艺条件:m(酶)∶m(仙人掌粉末)=1∶125,pH=4.5,酶解温度为55℃,酶解时间为2.0h,在该工艺条件下总皂苷平均提取率为0.182 5%。
Opuntia saponins is an important biologically active compone nt in ge nus apuntia ponins were extracted from opuntia dillenii by pectinase extraction with the extraction rate of saponins as the evaluated index , single factor test and orthogonal design were applied to investigate the effects of 4 factors including pH, the amount of enzyme , enzymolysis temperature and enzymolysis time. Ex-perimental results showe d that optimum extraction technology conditions were as follows: the mass ratio of enzyme and opuntia dillenii was 1 : 125,he pH was 4.5 ,enzymolysis temperature was 55 C , enzymolysis time was 2. 0 h,unde r these conditions,average extraction ratio of total saponins was 0.182 5 %.
