摘要
目的:观察真武汤对心力衰竭大鼠血清半乳糖凝集素-3(Galectin-3)及热休克蛋白70(HSP70)水平影响,探讨其治疗心力衰竭作用机制。方法:通过结扎大鼠冠状动脉前降支配合力竭式游泳成功复制心力衰竭大鼠模型,将造模成功的50只大鼠随机分成中药组(真武汤低、中、高剂量组)、西药组(卡托普利组)、模型组和空白组,每组10只,每日2次灌胃,持续15 d。用酶联免疫吸附试验法(ELISA)法测定大鼠血清BNP、Galectin-3及HSP70水平,并通过电镜观察心肌细胞超微结构。结果:真武汤组与西药组均可明显降低心力衰竭大鼠BNP水平,使Galectin-3及HSP70表达下调,真武汤低、中、高剂量组均优于或等同于西药组,电镜观察显示真武汤组和西药组心肌超微结构均有不同程度改善,真武汤大剂量组最为显著。结论:真武汤调控Galectin-3及HSP70的水平可能是其治疗心力衰竭的作用机制之一。
Objective: To observe the effects of Zhenwu Decoction on the Galactose lectin-3( Galectin-3) and Heat shock protein 70( HSP70) in the serum of rats with heart failure and explore the mechanism of the treatment of heart failure. Methods: By ligation of the rats,anterior descending coronary artery in combination with exhausted swimming,replicate rat model of heart failure successfully. Fifty successful model rats were randomly divided into Chinese medicine group( Zhenwu Decoction low,medium and high-dose groups),medicine group( captopril group),model group and blank group,10 rats in each group,twice a day orally for 15 days. Determine the levels of BNP,Galectin-3and HSP70 in rat serum by enzyme-linked immunosorbent assay( ELISA) and observe ultrastructural changes of myocardial cells by electron microscope. Results: Zhenwu Decoction groups and Western medicine group could reduce the level of BNP in heart failure rats significantly and lower the expressions of Galectin-3 and HSP70. Low,medium and high-dose Zhenwu Decoction groups were better than or equal to the western medicine group. The electron microscopy showed that myocardial ultrastructure was improved in Zhenwu Decoction group and Western medicine group and Zhenwu Decoction high-dose group was the most significant. Conclusion: Zhenwu Decoction in regulation of Galectin-3 and HSP70 levels may be one of the mechanisms in the treatment of heart failure.
