摘要
目的:研究鹿茸顶部组织中骨形态发生蛋白(BMPs)、转化生长因子-β(TGF-β)的分布,探索鹿茸生长发育机制。方法:采用免疫组织化学技术,观察和半定量分析了BMPs、TGF-β1在梅花鹿茸顶部组织细胞中的分布情况。结果:BMPs在表皮的基底细胞、毛囊细胞中均有强烈表达,真皮内的细胞、间充质细胞和前成软骨细胞无阳性反应,随着细胞的不断分化阳性细胞增加,软骨细胞有强阳性反应。从表皮向内,经真皮到间充质层,细胞外间质的BMPs棕黄色阳性反应渐渐减弱,平均总累计光密度值(IOD)由最高降至最低,从前成软骨层、过渡层到软骨层IOD又逐渐升高,但仍明显低于表皮层(P<0.01)。TGF-β1在各层均有分布,IOD值存在显著差异(P<0.05),从大到小按软骨层、过渡层、茸皮血管层、前成软骨层、间充质层、表皮层、毛囊层排列,血窦、血管和管膜形成区表达较强。结论:在梅花鹿茸顶部BMPs主要分布于表皮、毛囊和软骨层中,但表皮中的表达量最高,TGF-β1以不同的表达量分布于各层。
Objective: To study the distribution of bone morphogenetic proteins and transforming growth factor-β1 of antler tip tissue and explore their effects on antler growth and development mechanism. Methods: The distribution of BMPs and TGF-β1 in tip tissue of sika velvet antler were observed and semi quantitative analyse by immunohistochemical techniques. Results: BMPs were expressed strongly in epidermal basal cells,hair follicle cells,but not expressed in dermal cells,mesenchymal cells and prechondroblasts. As the cells differentiated,positive cells increased and cytoplasm had significant positive reaction. From the epidermis,through the dermis to the mesenchymal layer,BMPs brown positive reaction in the extracellular matrix was gradually weakened and the average integrated optical density( IOD) of BMPs was decreased from the highest to the lowest level. From the prechondroblasts layer,the transition layer to the cartilage layer,the IOD of BMPs increased gradually,but it was still significantly lower than that of the epidermis layer( P〈0. 01). TGF-β1 was distributed in every layers of tip tissue in sika deer antler. Their IOD values were significantly different( P〈0. 05) in different lays. The TGF-β1 expressions were stronger at the blood sinus,blood vessel and tube membrane formation regions. Conclusion: In tip tissue of sika velvet antler,BMPs mainly distributed in the epidermis,hair follicle and cartilage layer,but expressed the highest in the epidermis. TGF-β1 expressed differently in each layer.
