BCL10、Nramp1基因多态性与大白仔猪断奶前腹泻关联分析 被引量:3

Association between polymorphism of BCL10,Nramp1 gene and diarrhea traits in Large White piglets before weaning

摘要 为研究大白猪BCL10基因和Nramp1基因的单核苷酸多态性,并寻找与大白仔猪断奶前腹泻相关的分子标记,构建490头大白仔猪群体,对仔猪断奶前腹泻情况进行统计,利用PCR-RFLP法检测BCL10基因3′端1149T/C位点和Nramp1基因第六内含子NdeⅠ酶切位点多态性,同时与腹泻频率、腹泻指数和部分生长性状进行关联分析。结果显示:大白仔猪断奶前腹泻率为32.0%,重度腹泻个体的21日龄平均断奶重比正常个体低13.0%,日增重低16.2%,而且大白仔猪在7~11日龄和17~21日龄存在2个断奶前腹泻小高峰。仔猪腹泻粪便病原检测结果表明,22.5%的样本检出为沙门氏菌感染,17.5%的样本检出为致病性大肠杆菌感染。关联分析结果表明:BCL10基因多态性与初生重相关联(P<0.05),TT型个体初生重大于TC型个体,差异显著(P<0.05);BCL10基因多态性与腹泻频率、腹泻指数相关联,差异极显著(P<0.01),个体腹泻频率、腹泻指数均表现为TT>TC>CC,CC与TC之间差异显著(P<0.05),CC与TT之间差异极显著(P<0.01);Nramp1基因多态性与腹泻频率、腹泻指数相关联,差异极显著(P<0.01),个体腹泻频率、腹泻指数均为AA、AB型大于BB型,且AA型与BB型差异显著(P<0.05),AB型与BB型差异极显著(P<0.01)。研究结果表明,BCL10基因和Nramp1基因不同基因型对大白仔猪断奶前腹泻有显著影响,Nramp1基因与猪沙门氏菌感染导致的腹泻有关,BCL10基因3′端1149T/C位点和Nramp1基因第六内含子NdeⅠ酶切位点可以作为大白仔猪断奶前抗腹泻育种分子标记。 To explore the association between diarrhea of Large White piglets and polymorphism of BCLIO and Nrampll gene, a population of 490 Large White piglets before weaning was established and PCR-RFLP was carried out to examine the polymorphism of 3' 1149T/C locus of BCLIO gene and the Nde I restriction site in the 6th intron of Nrampl gene. Subsequently, correlation analysis of gene pol- ymorphism with diarrhea rate (DR), diarrhea index (DI) and several growth characteristics were performed. The results showed that DR of Large White piglets before weaning was 32.00//oo, average weaning weight and average daily gain of severe diarrhea individuals decreased by 13.0% and 16.2%, respectively. According to the results, piglets were easy to get diarrhea during two periods (7-11 and 17-21 days of age). Pathogen was detected from diarrhea piglets and 22.5% of them were caused by salmonella and 17.5 % by enteropathogenic Escherichia coli (EPEC). The polymorphism of BCL 10 gene was extremely significantly correlated with BW (P〈0.01). Specifically, DR and DI of individuals with different genotypes were TT〉TC〉CC, with difference between CC and TC was significant (P〈0.05) and between CC and TT was extremely significant (P〈0.01). The polymorphism of Nde I restriction sites in the 6th intron of Nrampl gene was extremely significantly correlated with DR and DI (P〈0.01) . The DR and DI level of piglets with AA and AB genotypes were much higher than that of BB genotype. The difference between AA and BB genotypes was significant (P〈0.05), and between AB and BB genotypes was extremely significant (P〈0.01). In summary, the result illustrated that polymorphism of BCL 10 and Nrampl gene was significantly related to diarrhea degree in Large White piglets, and the 3r 1149T/ C locus of BCLIO gene and the SNP of the Nde I restriction site in the 6th intron of Nrampl gene could be used as molecular markers for genetic selection in pig breeding.
