摘要
目的:研究金丝桃苷对D-半乳糖所致衰老模型小鼠的抗衰老作用。方法:采用D-半乳糖建立小鼠衰老模型,同时实验组分别灌胃不同剂量(25、50、100 mg/kg)的金丝桃苷,通过八臂迷宫试验和游泳耐力测验检测衰老模型小鼠短期学习记忆能力和抗疲劳能力。随后处死小鼠,采用ELISA法检测小鼠脑总抗氧化能力(T-AOC),超氧化物岐化酶(SOD)活性、谷胱甘肽过氧化物酶(GSH-Px)、活性氧(ROS)和丙二醛(MDA)含量,通过Western blot测定沉默信息调节因子1(Sirt1)、核因子-E2-相关因子(Nrf2)和醌氧化还原酶(NQO1)蛋白表达。结果:与模型组比较,金丝桃苷(25~100 mg/kg)小鼠游泳耐力更好,迷宫通过时间缩短、错误次数减少,脑组织T-AOC、SOD、GSH-Px活性明显升高,而脑组织ROS和MDA含量显著降低,并上调Sirt1、Nrf2和NQO1蛋白表达。结论:金丝桃苷对D-半乳糖致衰老小鼠具有抗氧化作用,其作用可能通过调控Sirt1/Nrf2信号通路相关。
Objective: To investigate Antioxidant effects of hyperoside in aging mice induced by D-galactose.Methods: D-galactose was used to establish aging model in mice,meanwhile,hyperoside( 25,50,100 mg/kg) were given in hyperoside groups for 42 days,then,eight arm maze experiment and swimming endurance experiment were performed,total antioxidant capacity( T-AOC),superoxide dismutase( SOD) activity,and glutathione peroxidase( GSH-Px),reactive oxygen( ROS) and malonaldehyde( MDA) in brain were determined by according ELISA kits,respectively.Sirtuin( Sirt1),nuclear factor erythroid-derived factor 2-related factor( Nrf2),quinone oxidoreductase( NQO1)protein expressions were detected by Western blot assay.Results: Compared with model group,hyperoside groups had longer swimming time,shorter time out of the maze and fewer numbers of error.The activities of T-AOC,SOD and GSH-Px in brains in hyperoside groups were markedly enhanced,and the ROS and MDA contents were decreased.Furthermore,hyperoside upregulated Sirt1,Nrf2 and NQO1 protein expression.Conclusion: Hyperoside has anti-oxidant and anti-aging effects,and the effects may be related to Sirt1/Nrf2 signaling pathway.
